宫颈组织中HPV16E6蛋白的表达

摘　　要：[目的]检测人乳头状瘤病毒(human papillomavirus,HPV)16E6蛋白在宫颈组织中的表达情况及其与宫颈病变的关系。[方法]选择宫颈脱落细胞学检查异常者中进行宫颈活组织检查的213例为研究对象,根据病理学诊断分为CCSC(宫颈鳞状细胞癌)组,共13例;CIN(宫颈上皮内瘤变)组,共114例;无病变组,86例。采用免疫印迹(Western blot)方法分别检测宫颈组织中HPV16 E6蛋白的表达。[结果]①宫颈脱落细胞学检查异常者CSCC发病率为6.1%(13/213),CINⅢ发病率为18.3%(39/213),CINⅡ发病率为23.9%(51/213),CINⅠ发病率为11.3%(24/213),无病变者占40.4%(86/213)。②3组宫颈组织中HPV16 E6表达阳性率分别为61.5%(8/13)、37.7%(43/114)、24.4%(21/86),两两比较P均﹤0.05。③相关分析发现HPV16 E6表达阳性率与宫颈病变严重程度呈正相关(r=0.68,P﹤0.05)。[结论]发生病变的宫颈组织中出现HPV16 E6阳性表达,其阳性率随病变级别的升高而增加。[Objective] To explore the relationship between HPV 16 infection and cervical dysplasia by examining the expression of human papillomavirus 16 （HPV16） E6 protein in cervical tissue. [ Methods] 213 patients who had abnormal cervical cell test accepted tissue biopsy. They were divided into 3 groups, cervical squamous cell carcinoma （CSCC） group, cervical intraepithelial neoplasia （CIN） group and no- neoplasia group by pathology diagnosis. All specimens were detected for HPV16 E6 protein, using Western blot techniques. [Results] ① Among 213 patients with abnormal cervical cell test, CSCC, CIN Ⅲ, CIN Ⅱand CIN Ⅰ incidence were 6.1% （13/213）, 18.3% （39/213）, 23.9% （51/213） and 11.3% （24/213） respectively.② The positive rates of HPV16 E6 protein in cervical tissues in 3 groups were 61.5% （8/13）. 37.7% （43/114）. 24.4% （21 /86） respectively （P 〈 0.05 for all）. ③ Correlation analysis showed that the positive rate of HPV16 E6 protein had positive correlation with severity of cervical lesions （r = 0.68. P 〈 0.05）. [Conclusion ] The development of cervical dysplasia may be associated with HPV16 infection. The more serious the cervical lesions are, the more higher the positive rate of HPV16 E6 is.
