槲芪散及其君药槲寄生对大鼠肝癌前病变组织中促凋亡因子Omi/HtrA2表达的影响

摘　　要：目的:探讨槲芪散及其君药槲寄生干预肝癌前病变组织中癌细胞凋亡机制。方法:用电镜观察模型组、正常组以及槲芪散大小剂量组、槲寄生单体组肝细胞线粒体形态学的变化,用免疫组化SP法、Western Blotting法检测各实验组大鼠肝组织丝氨酸蛋白酶Omi/HtrA2的表达。结果:槲芪散治疗组肝细胞线粒体形态较模型组有明显改善;除正常组外,模型组,槲芪散大、小剂量治疗组,槲寄生单体组均有Omi/HtrA2的表达,模型组、正常组、大剂量治疗组、单体组之间丝氨酸蛋白酶的表达量有统计学意义(P<0.05)。结论:槲芪散及其君药槲寄生能够改善肝细胞线粒体的形态和功能,降低Omi/HtrA2总量的表达,并且促使Omi/HtrA2从线粒体释放,诱导肝癌细胞的凋亡。Objective： To investigate the apoptosis mechanism of the serine protease Omi/HtrA2 in rat lesion precancerous by Huqi powder and its principal drug mistletoe extracts. Methods： Mitochondria morphology was observed by electron micro- scope in model group, Huqi powder great and small dose group, normal group. The expression of Omi/ HtrA2 was assayed by immunohistochemcal means and Western Blotting in model group, Huqi powder great and small dose group, mistletoe extracts group, normal group. Results： Mtochondrial morphology in the treated group of rat is significant improvement by comparing with the model group. Except the normal group, the other groups all expressed positive Omi/HtrA2. The difference among the model group, the normal group, the high-dose treatment group and the mistletoe extracts group was of significance in statistics science （ P 〈 0. 05 ） . Conclusion： Huqi powder and principal drug mistletoe extracts can improve mitochondria 1 morphology and function, reduce the total expression Omi/HtrA2, spur Omi/HtrA2 release from the mitochondria and induce apoptosis in cancer cells.
