摘  要:目的建立一种人脑膜瘤细胞原代培养简单有效的方法。方法新鲜脑膜瘤组织取自福建协和医院神经外科25例连续治疗的脑膜瘤手术病人,以胰蛋白酶消化法分离细胞,用含10%胎牛血清的DMEM培养液进行培养并传代,显微镜下观察脑膜瘤细胞生长状况。上皮膜抗原(EMA)、波形蛋白(Vimentin)对脑膜瘤细胞进行免疫组化鉴定。结果用胰酶消化法培养脑膜瘤细胞,方法简单,细胞形态典型;EMA、Vimentin表达阳性,鉴定为脑膜瘤细胞。结论单一的胰蛋白酶消化法是一种简单有效的脑膜瘤细胞原代培养的方法。Obiective To develope a simple and effective method of culturing primary human meningioma cells. Methods Tumor specimens were obtained from 25 consecutively treated adult patients who underwent surgery for resection of intracranial meningiomas in the Union Hospital of Fujian. Fresh meningioma specimens were cut into small pieces and digested with 0. 25% trypsin. Cell suspension was cultured in DMEM containing 10% fetal bovide serum (FBS). Cultured cells were characterized by microscope. Cells were analyzed for immunophenotyping with the following antibodies: vimentin, epithelial membrane antigen (EMA). Results The appearance of meningiomas cell is typical and the method is simple. The cultured meningioma cells retained the main feature of the primary tumor, being positive both for vimentin antigens and for EMA. Conclusions Trypsin digestion is simple and effective for isolation of meningioma cells.
