摘 要:目的探讨人脐血干细胞(HUCB-SCs)蛛网膜下腔移植治疗中枢神经系统损伤患者在体内向神经干细胞(NSCs)分化的状态。方法采用流式细胞术检测HUCB-SCs蛛网膜下腔移植前后神经干细胞特异性标志物巢蛋白(Nestin)表达变化。结果HUCB-SCs蛛网膜下腔移植1周后,脑脊液中原始幼稚/干细胞Nestin抗原荧光表达明显增强,Nestin阳性细胞占原始幼稚/干细胞比例为(7.58±6.43)%,明显高于HUCB-SCs中Nestin阳性细胞比例(P<0.05)。结论HUCB-SCs蛛网膜下腔移植后,在适当条件下在体内可以向神经干细胞分化,为脐血干细胞移植用于神经系统疾病的临床治疗提供较直接的客观依据。Objective To investigate the differentiation of human umbilical cord blood stem cells (HUCB-SCs) into neural stem cells (NSCs) after transplanting into the subarachnoid space in the treatment of patients with central nervous system injury. Methods The expression of Nestin, a neurofilament protein as one of the most specific markers of neural stem cells, on HUCB-SCs before and after transplantation was detected by applying flow cytometry. Results The antigen fluorescent expression of Nestin was obviously enhanced on the progenitor or stem cells in cerebrospinal fluid one week after HUCB-SCs transplantation. The Nestin-positive cell ratio in the progenitor or stem cells in cerebrospinal fluid was about (7. 58±6.43)%, significantly higher than that of HUCB-SCs (P〈 0.05). Conclusion These data suggest that HUCB-SCs may differentiate into NSCs under proper conditions in vivo. Our study provides the directed objective basis for HUCB-SCs transplantation in the clinical treatment of nervous system diseases.
