摘 要:目的观察大鼠液化石油气中毒后脑细胞形态改变及胶质纤维酸性蛋白(GFAP)表达的变化。方法采用大鼠液化石油气中毒模型,动物分别于中毒后1、3、7、14、21、28d处死,苏木精-伊红染色观察脑组织神经细胞的动态变化,免疫组化方法检测脑组织GFAP的表达水平。结果大鼠液化石油气中毒后大脑皮质神经元细胞核浓缩、结构不清,随中毒浓度增加,细胞核浓缩更明显,但未见明显神经元坏死迹象。免疫组织化学技术观察显示GFAP表达增加,随中毒浓度升高,GFAP表达亦明显增加。结论液化石油气中毒后大脑皮质神经元形态学改变是认知功能下降的病理基础之一,脑组织GFAP表达的增加是损伤修复的一种表现。Objective To observe the changes of morphology of neurocyte and the expression level of GFAP in rats after acute liquid petroleum gas poisoning. Methods Liquid petroleum gas poisoning model of rats was used at 1,3,7,14,21 and 28 d after poisoning, rats were killed respectively. The morphological changes of neurocyte were observed by HE staining. The expression levels of GFAP in brain were detected by immunohistochemistry. Results The degeneration of neurons and inspissation of karyon were observed in LPG treated rats, and the expression of glial fibrillary acidic protein increased with the gliosis in response to LPG treatment. Conclusion The changes of morphology of neurocyte after acute liquid petroleum gas poisoning is one of the mechanisms of cognitive dysfunction. The expression of GFAP increased in brain is an injury repair phenomenon.
