摘 要:目的动态观察丹酚酸B(SalB)对脑缺血再灌注小鼠血管内皮细胞分泌功能的影响。方法NIH小鼠随机分为假手术组、模型组、丹酚酸B治疗组和尼莫地平对照组,复制脑缺血再灌注病理模型,并分别在脑缺血30 min再灌注0、30、60 min时测定脑组织一氧化氮(NO)、血浆内皮素(ET)、组织型纤溶酶原激活物(t-PA)、组织型纤溶酶原激活物抑制剂(PAI-1)以及血管性假血友病因子(vWF)的含量。结果模型组不同时点的ET,NO,PAI-1和vWF含量明显升高,而t-PA的含量降低。用丹酚酸B治疗,可调整内皮细胞的分泌功能,尤其是再灌注60 min时可显著降低ET、NO、PAI-1和vWF的含量(P(0.01或P<0.05),同时升高t-PA的含量(P(0.01)。结论丹酚酸B可能通过调整血管内皮细胞分泌舒缩、促凝和抗凝物质的平衡,起到减轻缺血再灌注脑损伤的作用。Objective To observe dynamically the influence of salvianolic acid B(SalB)on secretive function of vascular endothelial cells in mice with cerebral ischemia reperfusion.Methods NIH mice were randomly divided into sham-operated group,model group,SalB-treated group and Nimodipine-collated group.The model with cerebral ischemia-reperfusion was established.After ischemia for 30 min followed by 0 min,30 min,60 min of reperfusion,Nitrogen monoxidum(NO) content from brain tissue was determined,and the contents of endothelin(ET),tissue-type plasminogen activator(t-PA),plasminogen activator inhibitor-1(PAI-1) and von Wilebrand factor(vWF) in blood plasm were measured respectively.Results At diefferent time after cerebral ischemia reperfusion injury,contents of ET,NO,PAI-1 and vWF improved remarkably,whereas t-PA content decreased in the model group compared with the sham-operated group.However,SalB regulated secretive function of endothelial cells,especially at 60 min after reperfusion.It decreased markedly contents of ET,NO,PAI-1 and vWF(P﹤0.01 or P﹤0.05),and improved content of t-PA(P﹤0.01).Conclusion SalB can protect cerebral ischemia reperfusion from injury,which may contribute to its action on regulating the balance among dilatation,contraction,procoagulant and anticoagulant substances secreted by vascular endothelial cells.
