摘 要:目的研究辅酶Q10对紫外线所致角膜上皮细胞光损伤防护作用。方法36只Wistar雄性大鼠随机分为正常对照组、阳性对照组与辅酶Q10治疗组。紫外线连续照射24 h建立角膜上皮紫外线光损伤模型。治疗组于光照前3周每天辅酶Q10灌胃。观察角膜组织病理学改变并用免疫组化染色观察凋亡相关基因蛋白Bcl-2、Bax、Fas和FasL的表达,进行细胞计数与统计学检验。结果正常对照组角膜上皮可以见到Bcl-2、Fas和FasL的阳性表达,很少见到Bax的阳性表达。阳性对照组和辅酶Q10治疗组均可以见到Bcl-2、Bax、Fas和FasL的表达。与正常组比较,阳性组Bcl-2表达阳性的角膜上皮细胞数显著减少,Bax表达阳性的角膜上皮细胞数显著增多(P<0.01);治疗组Bcl-2、Bax表达阳性的细胞数与正常组相比无明显变化(P>0.05);而阳性组Fas和FasL表达阳性的角膜上皮细胞数明显增多(P<0.01),治疗组Fas和FasL表达阳性细胞数明显少于阳性组(P<0.05)。结论辅酶Q10通过上调Bcl-2的表达,下调Bax、Fas和FasL的表达,能够阻抑凋亡过程和细胞的死亡。Objective To explore the protective effect of coenzyme Q10 on corneal epithelium of UV irradiation-induced injury rats and its mechanisms. Methods Thirty-six male Wistar rats were divided randomly into normal control group, positive control group and coenzyme Ql0 group. The UV-induced damage model of Wistar rats was established by exposing to ultraviolet for 24 hours. The rats in coenzyme Q10 group were intragastric administrated by coenzyme Q10 everyday for three weeks before UV exposure in different group. The rats in normal group did not received ultraviolet light, and the rats from positive group only underwent uhraviolet ray. without the administration of drug. The Changes of corneas histopathology were observed and the expressions of Bcl -2, Bax, Fas and Fasl were observedby immunohistochemistry . Cell counts andr statistical analysis were used to assess results. Results Bcl -2, Fas and Fasl were found in normal group and Bax were hardly found in normal group. Bcl-2, Bax, Fas and Fasl were observed in positive control group and coenzyme Q10 group. Compared with nor- mal group, the expression of Bcl - 2 were obviously decreased and the expression of Bax were obviously increased in positive control group (P 〈 0. 01 ) ; The cells expressing Bcl - 2 and Bax in coen- zyme QIO group had no change ( P 〉0. 05 ) There was increase of expression amount of Fas and Fasl in positive group (P 〈 0. 01 ). It was also found that cells of expressing Fas and Fasl in coenzyme Q10 groupwere obviously less than that in positive group (P 〈 0.05). Conclusions Coenzyme Q10 can up-regulate Bcl -2 expression and down-regulate Bax, Fas and Fasl expression and inhlbit apoptosis of cells in corneal epithelium.
