摘 要:目的探讨钌红在脂多糖(LPS)致大鼠发热过程中对体温的影响及可能的机制。方法用LPS复制大鼠发热模型,观察不同浓度钌红对LPS发热曲线的影响及发热时下丘脑组织中TRPV4表达的变化。结果(1)与单独给予LPS组相比,随着给予钌红浓度的增大,LPS致发热幅度均有不同程度的降低;(2)单独给予钌红与给予生理盐水的对照组比较,体温有所降低;(3)钌红组及钌红+LPS组TRPV4表达明显低于对照组。结论(1)钌红对LPS致热性有抑制作用;(2)钌红能调节TRPV4通道的表达;(3)TRPV4通道可能参与正常体温的维持。Objective To observe the effect of ruthenium red (RR) on the body temperature of rats with lipopolysaccharide (LPS)-induced fever and investigate the possible mechanism. Methods Rat models of fever were established with lipopolysaccharide and the effects of RR at different doses were observed on the body temperature of the rats and the content of TRPV4 in the hypothalamus. Result Compared with those in LPS group, the rats with LPS-induced fever receiving RR treatment showed a dose-dependent lowering of the body temperature. The rats with RR treatment had lower body temperature than those with saline injection. The content of TRPV4 in the saline group was significantly higher than that in RR+LPS and RR group. Conclusion RR inhibits LPS-induced fever in rats and regulates the hypothalamal expression of TRPV4 channels, which may participate in the maintenance of normal body temperature.
