摘 要:目的观察二亚硝基哌嗪在正常状态和气虚状态大鼠鼻咽上皮细胞的诱癌效率差异以及黄芪、黄连复方有效成分的防护作用。方法SD大鼠每日力竭游泳10min,连续30d以建立气虚状态模型,然后分组,每组动物10只。DNP皮下注射法诱癌,并应用黄芪、黄连复方有效成分进行干预。干预完成后200天,分别取各组动物鼻咽黏膜行常规病理组织学和超微病理观察。结果力竭游泳联合DNP诱癌组动物中,鼻咽黏膜上皮细胞出现癌前病变例数(n1=8)显著多于单纯DNP诱癌组(n2=3,P<0.05),而应用益气解毒中药干预后,该类动物鼻咽黏膜上皮细胞癌前病变例数显著降低(n3=3,P<0.05)。结论气虚状态下,实验动物鼻咽黏膜上皮细胞对DNP诱癌效应更加敏感,而黄芪、黄连复方活性成分能够有效阻逆该类病理过程。Objective To observe the differences in the occurring rates of induced precancerous lesion by DNP (N, N'-dinitrosopiperazine) in nasopharyngeal epithelia among rats either at a normal physique status or at a Qi deficient one and to investigate the inhibitory effects of active components of milkvetch root (MR) and Compound Coptis Root Formula (CCRF) on the development of this kind of induced precancerous lesion. Methods Included in this experimental study were 50 rats, to prepare the physique model of Qi deficient status at first by a procedure of 10 min of forced exhaustedly swimming each day and lasted for a continuing period of 30 days. Then, the animals were divided into several groups for the following experimental procedures, with 10 rats in each group. Followed was the carcinogenesis-inducing program to prepare the model of precancerous lesion in the nasopharynx by use of DNP injected subcutaneously at a dosage of 0.5 mg each time for each animal, twice one week and 30 times in total. Meanwhile, those rats in the treating groups were given the active components of MR and CCRF to intervene the progress of carcinogenesis-inducing lasted for 30 days respectively. Two hundred days late following the end of intervening program, all these animals were put into death to collect mucosal tissue samples from the nasopharynx for routine and ultrastructural pathohistological observation. Results In the group of rats undergone forced exhaustedly swimming and carcinogenesis-inducing procedure by DNP, the number of animals with precancerous lesion in the nasopharyngeal mucosa was significantly higher (n1=8) than that of animals with a carcinogenesis-inducing procedure simply by DNP injection (n2=3,P〈0.05). In contrast, this number was significantly lowered (n3=3,P〈0.05) in the animals of groups received the intervening program composed of the active components of MR and CCRF as compared with that of modeling groups.
