摘 要:目的研究芦笋多糖对S180小鼠红细胞膜带3蛋白Cl-转运功能的影响及其机制。方法采用荧光分光光度法动态观察红细胞内Cl-的含量;应用流式细胞仪测定红细胞膜电位大小。结果在Cl-转运功能实验中,模型对照组在20,35min的红细胞荧光强度均明显低于正常组,芦笋多糖3个剂量组红细胞荧光强度较模型对照组显著升高,低、中剂量组作用非常显著(P<0.01),高剂量组在35min也有显著作用(P<0.05);在膜电位实验中,模型对照组S180小鼠荧光强度(45.0%)较正常小鼠(99.4%)显著降低(P<0.01),而低、中、高剂量芦笋多糖能使S180小鼠红细胞膜电位显著升高(P<0.01)。结论芦笋多糖能够显著增强红细胞膜的Cl-转运的功能,并且芦笋多糖的这一作用可能是通过升高红细胞膜电位而实现的。OBJECTIVE To observe the influence of Asparagus polysaccharide on CI - transportation function of band 3 protein on the erythrocyte membrane of S180 mice and its mechanism. METHODS Cl^- transportation function was explored by the fluorescence spectrophotometric method, and Flow Cytometery (FCM) was used to examine the erythrocyte membrane potential. RESULTS In the examination of Cl^- transportation, the fluorescence intensity of negative control group was remarkably lower than that of the normal group, and the fluorescence intensity of polysacchar/de groups was higher than that of negative control group, the low dose group and the medium dose group ( P 〈 0. 01 ), the high dose group ( P 〈 0. 05 ) at 35 min after reaction. In the experiment of membrane potential, the fluorescence intensity of negative group(45.0% ) was remarkably lower than that of the normal group(99. 4% ) , and the fluorescence intensity of polysaccharide groups (65.5% , 98. 4% , 89.9% ) was higher than that of negative group. CONCLUSION Asparagus polysaccharide obviously enhanced the C1^- transportation function of erythrocyte band 3 of S180 mice, and the action was obtained possibly through elevating erythrocyte membrane potential.
