摘  要:目的观察环氧酶-2(COX-2)、基质金属蛋白酶-3(MMP-3)和组织特异性抑制物-1(TIMP-1)在骨性关节炎(OA)中的表达,探讨其与OA的发生、发展的关系。方法采用苏木精-伊红染色及SABC法,检测OA及正常对照关节软骨组织切片中COX-2、MMP-3和TIMP-1蛋白的表达情况,应用统计学方法对其在OA的发生、发展中与OA病理分级、临床影像学分级的相关性进行非参数分析。并运用Spearman相关分析检验对OA组中MMP-3蛋白与COX-2蛋白的表达进行相关性分析。结果OA中MMP-3、TIMP-1、COX-2蛋白的阳性表达率分别为78.0,68.7和86.7,与它们在正常软骨组织中的比较有显着性差异(P<0.05),MMP-3、COX-2与关节损伤程度成正相关(P<0.05),TIMP-1与关节损伤程度成负相关(<0.05)。结论OA中MMP-3/TIMP-1的变化与COX-2变化无相关性。Objective To investigate the role of cyclooxygenase 2 (COX- 2), matrix metalloproteinase 3 ( MMP- 3) and tissue inhibitor of metalloproteinase 1 (TIMP- 1) in osteoarthritis (OA) by observing COX - 2, MMP- 3 and TIMP - 1 protein in OA, and to provide objective prediction index for OA' s clinical biological behaviors, clinical diagnosis and therapeutic methods selection. Methods The expression of COX- 2, MMP- 3 and TIMP - 1 was detected by immunohistochemical ultrasensitiveTMS- P method from paraffin embedded specimens of osteoarthritis tissues and normal cartilages tissues. Correlation was studied by analysis of clinical biological behaviors of OA and statistical method. Results In osteoarthritis tissues, the positive expression rate of COX - 2, MMP - 3 and TIMP- 1 were 78.0%, 68.7% and 86.7% respectively, which was significantly higher than normal cartilages specimens (P 〈 0.05). Significant differences of the expression rates of COX- 2, MMP- 3 and TIMP- 1 were found between different degrees of OA (P〈0.05). In the Manki stage, the positive expression rate of COX-2, MMP-3 increased significantly from I to H , then to m (P〈 o. 05). In osteoarthritis tissues, there was no significant correlation between COX- 2 and MMP- 3. Conclusion There is no interaction between MMPs TIMPs and COX- 2 in the development of OA.
