摘 要:目的探讨温热联合顺铂(DDP)对人卵巢浆液性乳头状囊腺癌细胞株SKOV3的毒性作用及量效关系。方法以体外培养的人卵巢浆液性乳头状囊腺癌细胞株SKOV3为研究对象,采用配伍设计的方法,恒温水浴加热,以四甲基偶氮唑蓝(MTT)比色法检测不同加热温度、加热时间、药物浓度对SKOV3的细胞毒作用的影响。结果(1)加热时间设为30min,不同浓度DDP的细胞毒指数(CI)值均随加热温度的升高而逐渐增高,40℃以上各不同浓度DDP的CI值与37℃时比较,差异有统计学意义(P<0.05),且温度越高,差异越显著(P<0.01)。(2)加热温度固定为43℃,不同浓度DDP的CI值均随加热时间的延长而逐渐增高,加热30min时与15min以内各时间的CI值差异均有统计学意义(均P<0.05);加热60min时与30min以内各时间的CI值之间差异更显著(均P<0.01),但加热15min以内时的CI值差异无统计学意义(P>0.05)。结论增高加热温度或延长加热时间均可以显著提高DDP对SKOV3的细胞毒作用。Objective To investigate the cytotoxic effect of hyperthermia combined with chemotherapy of cisplatin on human ovarian carcinoma SKOV3 cells. Methods The cultured humar) ovarian carcinoma SKOV3 cells were treated with different concentrations of cisplatin (0ug/ml, 0.1ug/ml, 1ug/ml, 10ug/ml, 100ug/ml), different temperatures (37.0℃, 40.0℃, 43.0℃, 45.0℃) and different times (0 min, 7.5 min, 15 min, 30 min, 60 min). The cytotoxic effect was assessed by MTT method. Results When the hyperthermic time was 30 minutes, the cytotoxicity index (CI) of each group went higher with the elevated temperature; the CI of temperature 〉40~C was significantly higher than that of 37℃( P 〈 0.05). When the temperature was 43℃, the CI of each group went higher with the time prolonged, the CI of hyperthermia 〉30 min was significantly than those 〈15 min ( P〈 0.05 ), the CI of hyperthermia with 60 rain was significantly higher than those with 〈30 min (P〈0.01); but when the hyperthermic time 〈 15 min there was no significant difference in CI among all groups (P〉 0.05). Conclusion The cytotoxic effect of cisplatin on SKOV3 cells can be enhanced by prolonged hyperthermic time or elevated temperature.
