摘 要:分离了1株溴氨酸降解菌,其可以溴氨酸为唯一碳源进行降解并使其脱色,通过16S rRNA基因序列比较和生理生化特性分析,将其归为鞘氨醇单胞菌属.溴氨酸降解和菌株生长的最适条件为:温度30℃,pH 7.0,摇床转速100 r/min,(NH4)2SO4作为氮源,在此条件下,溴氨酸(100 mg/L)在14 h内的脱色率可达99%.低浓度NaCl(<2%)对脱色有促进作用,而高浓度NaCl(≥2%)对脱色产生抑制.以Haldane底物抑制模型表征溴氨酸初始浓度对脱色的影响,确定当初始浓度为1 393.5 mg/L时可取得最佳比降解速率1.4 h-1.菌株不能将溴氨酸完全矿化,至反应终点52.4%的有机碳得到去除.利用GC-MS和HPLC-MS分析代谢产物显示,溴氨酸降解的中间产物是邻苯二甲酸,终产物可能为2-氨基-3-羟基-5-溴苯磺酸或2-氨基-4-羟基-5-溴苯磺酸,邻苯二甲酸可经3,4-二羟基苯甲酸途径进一步降解而被菌体利用.A bacterial strain that could degrade bromoamine acid (BAA) as the sole carbon source was isolated. It was identified as Sphingomonas sp. based on 16S rRNA gene sequence analysis and physio-biochemical characteristics. Under the optimal growth conditions, with temperature of 30℃ , pH of 7.0, rotating rate of 100 r/min and (NH4)5 SO4 as the nitrogen source, the decolorization percentage of BAA (100 mg/L) could reach 99% within 14 h. NaCI of low concentration ( 〈 2% ) facilitated the decolorization, while NaCI of higher concentration ( 1〉 2% ) had inhibition effect. The effect of initial BAA concentration on decolorlzaiton could be described by the Haldane model, and the optimal specific decolorlzation rate of 1.4 h-r could be obtained when the initial concentration of BAA was 1 393.5 mg/L. The strain could not mineralize BAA completely, and 52.4% of the total organic carbon was removed at the end of the reaction. The analysis of metabolites using GC-MS and HPLC-MS showed that phthalic acid was the metabolic intermediate which could be further degraded through 3, 4-dihydroxybenzoic acid route and serve as the growth substrate, and the end product was estimated to be either 2-amino-3-hydroxy-5- bromobenzenesuffonic acid or 2-amino-4-hydroxy-5-bromobenzenesulfonic acid.
