摘 要:目的观察铅、乙醇染毒对雄性小鼠初级精母细胞致突变性的影响,研究铅和乙醇对雄性小鼠联合生殖毒性。方法健康雄性小鼠,随机分为9组,每组8只。A~H组经灌胃分别给予醋酸铅15mg/kg体质量、30mg/kg体质量,乙醇1g/kg体质量、2g/kg体质量,醋酸铅15mg/kg体质量+乙醇1g/kg体质量,醋酸铅15mg/kg体质量+乙醇2g/kg体质量,醋酸铅30mg/kg体质量+乙醇1g/kg体质量,醋酸铅30mg/kg体质量+乙醇2g/kg体质量,对照组给等量蒸馏水,1次/d,每周5次,连续3周。所有动物均于染毒第25d处死,取双侧睾丸作初级精母细胞染色体畸变分析。结果高铅染毒组、高乙醇染毒组、铅和乙醇联合染毒组初级精母细胞染色体畸变率与对照组相比显著增加(P<0.05),析因分析表明醋酸铅和乙醇之间存在交互作用。结论铅和乙醇对于雄性小鼠初级精母细胞具有致突变性,并且存在协同作用。Objective To observe the mutagenesis in male mice primary spermatocytes induced by lead and ethanol separately or Combinationally. To clarify whether lead and ethanol have combined effect on male mutation.: Methods Healthy male mice were divided into A~ H groups randomly with eight in each. The mice in treated groups were gavaged respectively with lead acetate at 15,30 mg/kg body weight(BW),ethanol at 1,2 g/kg BW,lead acetate at 15 mg/kg BW+ ethanol at 1 g/kg BW,lead acetate at 15 mg/kg BW+ ethanol at 2 g/kg BW,lead acetate at 30 mg/kg BW+ ethanoi at 1 g/kg BW,lead acetate at 30 mg/kg BW+ ethanol at 2 g/kg BW for once a day, 5 times a week, lasting three weeks. The mice in control group were gaved with distilled in equal volume and a same treated way. At the 25th days after first exposure,all the mice were killed and the testicle were took out to find primary spermatocyte chromosome aberration (RPSCA). Results The rate of RPSCA of groups treated with high lead acetate,high ethanol,and lead acetate+ethanol was much higher than that of control( P 〈0.01). Factorial analysis showed an interaction between lead acetate and ethanol in inducing mutagenesis in male mice primary spermatocyte. Conclusion Lead and ethanol induced the mutagenesis on male mice primary spermatocytes respectively,and the synergetic effect between lead and ethanol was observed.
