XAGE-1b腺病毒表达载体的构建及其功能研究

摘　　要：XAGE是通过生物信息学方法发现的新一类睾丸—肿瘤抗原(cancer/testis antigen,CT抗原)基因,XAGE-1是其主要成员.当前已获得了XAGE-1在多种肿瘤组织中较为详细的表达谱,并初步揭示了XAGE-1与病理进程及肿瘤亚型的关系,但关于其功能及表达调控机制的研究尚刚刚起步.构建了高效表达XAGE-1b的重组腺病毒载体Adv-1b;在细胞增殖、集落形成实验中,感染Adv-1b后的细胞增殖能力更强(P<0.01,studentt检验),形成的单克隆集落更大;软琼脂集落形成实验中,感染Adv-1b后的细胞集落形成率更高(P<0.01,卡方检验).研究结果初步证实:XAGE-1b对涎腺腺样囊性癌细胞的增殖及转移能力有一定的促进作用,为进一步深入研究XAGE-1b的功能和致癌机制打下了良好基础.XAGE discovered by bioinformatics, is a new type of cancer/testis antigen （ CT antigen）, whose main member is XAGE-1. Now a relatively precise expression profile of XAGE-1 in cancer tissues and the correlation between XAGE-1 expression and the subtypes or the stages of cancers have been known, but its function and expressing regulation is unlinown. Here, two recombinant adenovirus strains were constructed, one of which could highly express XAGE-1b, named Adv-1b. Compared with control cells, cells infected with Adv-1b showed higher multiplication rate by cell multiplication assay （P〈0.01, student t test） ,and formed bigger cell colonies by colony formation assay and more cell colonies by soft agar colony formation assay （P 〈 0.01, Х^2 test）. From the results, we conjectured XAGE-1b could promote the multiplication and metastasis of cancer cells. These findings are significant for further study on XAGE-1b＇ s function and role in tumor occurrence.
