ESTABLISHMENT OF RETINAL ISCHEMIA-REPERFUSION MODEL WITH THREAD OCCLUSION IN RAT:A PRELIMINARY STUDY

摘  要:目的建立一种理想的视网膜缺血再灌注动物模型。方法成年Wistar大鼠20只,除2只4眼作为对照组外,余18只36眼作为实验组,分为缺血1 h组及缺血2 h组,每组9只。实验中用预先制备的线栓插入实验组大鼠颈内动脉,造成视网膜缺血,分别于缺血12、h后及再灌注6 h后观察眼底,术后摘除缺血1、2 h及再灌注6 h的眼球进行组织学观察。结果实验组大鼠术后均发生视网膜缺血,视网膜苍白、水肿,以神经纤维层和内网状层最明显,可见白细胞浸润。再灌注6 h后视网膜水肿减退,神经纤维层水肿减轻,造模成功。结论此方法比较接近视网膜缺血临床病理过程,模型稳定、可靠。Objective To establish an ideal retinal ischemia-reperfusion animal model. Methods A segment of pre pared thread was introduced into the carotid artery of 18 adult Wistar rats and resulted in retinal ischemia of 36 eyes, which was followed by 6 h reperfusion. Fundus changes and morphological changes of eyes were observed after ischemia and reperfusion. Ac cording to the period of ischemia, the 36 eyes were divided into two groups: group 1, having 1 h ischemia, group 2, having 2 h is- chemia. Four eyes of two rats served as controls. Results The retinal ischemia resulted in retinal pallor and edema with leuko cytic infiltrate, especially in nerve fiber layer and plexiform layer. After reperfusion, retinal edema and nerve fiber layer edema be- gan to decrease. Conclusion The retinal ischemia model established in this study is similar to the natural course of clinical retinal ocular ischemia. The model is stable and reliable.
