前列腺癌和前列腺增生症中诱导型一氧化氮合酶的表达

摘  要:目的测定诱导型一氧化氮合酶在正常前列腺、前列腺增生症、前列腺癌三者中及前列腺癌不同病理分级、临床分期中的表达情况。方法应用链霉菌素抗生物蛋白-过氧化酶方法,分别用诱导型一氧化氮合酶多克隆抗体对正常前列腺、前列腺增生症、前列腺癌行免疫组织化学染色,采用免疫组织化学染色评分方法对诱导型一氧化氮合酶表达阳性切片行定性评分。结果诱导型一氧化氮合酶在正常前列腺几乎无表达,在前列腺增生症低表达,在前列腺癌高表达,三者差异具有显著性(P<0.01);但诱导型一氧化氮合酶在不同病理分级、临床分期前列腺癌中的表达强度差异无显著性(P>0.05)。结论诱导型一氧化氮合酶在正常前列腺无表达、在前列腺增生症低表达、在前列腺癌高表达,表明其与前列腺增生症和前列腺癌的发生有关。Objective To evaluate the expression of inducible nitric oxide synthase (iNOS) in normal prostate (NP), benign prostatic hyperplasia (BPH), and prostate cancer(PC), as well as in different grading of pathological and clinical system. The aim of our work is to explore the role and significance of iNOS in the development of BPH and PC. Methods According to pathological grade, PC were divided into three groups: 16 high-grade, 19 moderate-grade, 15 low-grade. According to clinical stage, PC were divided into four groups: 12 A, 14 B,13 C, 11D. Expression of iNOS was detemined by immunohistoehemical staining. Polyclonal antibody against iNOS was used for qualitative identification. Results The immunohistoehemical staining of iNOS in NP, BPH and PC were negative, weak and highly positive respectively. The differences were statistically significant (P 〈 0.01). While within the specimens of PC group no significant differences were noted among various pathological grades and clinical stages (P〉0.05). Conclusions Our study indicated that prostatic hyperplasia and malignant changes were closely associated with the intensity of iNOS.
