摘 要:目的探讨p53、MIF(macrophagemigration inhibitory factor)在前列腺癌(prostate cancer,Pca)中的发生、发展的作用,为前列腺癌的诊断及治疗提供新途径。方法采用免疫组化方法检测60例良性前列腺增生症(BPH)和58例前列腺癌(Pca)组织中MIF、p53蛋白的表达。结果Pca组织p53与MIF阳性表达率分别为48.28%(28/58)和79.31%(46/58),均明显高于BPH组(P<0.05);p53与MIF表达呈明显正相关(P<0.05);两者在Pca组织中的表达水平与其病理分级和临床分期均呈正相关(P<0.05);p53和MIF阳性表达的肿瘤复发快、易转移、预后差。结论p53和MIF与Pca组织学分级、恶性程度和预后密切相关,是检测Pca的较好分子标志物;联合检测p53和MIF的表达可作为判断Pca生物学行为及预后的重要参考价值。Objective To discuss the significance of p53 and MIF in carcinogenesis, progression and angiogenesis of prostate carcinoma, providing an approach for the diagnosis and treatment of the tumor. Methods The expression of MIF and p53 in 60 benign prostate hyperplasia(BPH) and 58 prostate carcinoma(Pca)was examined with immunohistochemistry. Results The positive expression rates of p53 and MIF in58 cases of Pca were 48.28% and 79.31%, respectively. The expression levels of p53 and MIF in prostatic carcinoma were much higher than those in BPH (P〈0.05) ;the positive correlation was noted between p53 expression and MIF expression(P〈0.05) ;and both of them were significantly related to the tumor pathologic grades(P〈0.05) and tumor clinical stage (P〈0.05. Tumors with high levels of p53 and MIF positive recurred earlier and had a greater risk of stage progression than those with low levels of p53 and MIF negative. Conclusion This study indicates that p53 and MIF are closely related to the hist opathological grading and the tumor differentiation and prognosis of Pca.p53 and MIF are good molecular markers of Pca. The combined detection of p53 and MIF may serve as an important referrence to estimate the biological behavior and prognosis of Pca.
