红花黄色素粉针对肾间质纤维化实验大鼠Ⅲ型胶原及转化生长因子-β_1的影响

摘　　要：目的观察红花黄色素粉针对肾间质纤维化实验大鼠Ⅲ型胶原(collagenⅢ,ColⅢ)及转化生长因子-β1(transforming growthfactor-1,TGFβ1)的影响。方法选用健康、雌性SD大鼠40只,用单侧输尿管结扎(unilateral ureteral obstruction,UUO)方法复制大鼠肾间质纤维化模型,将其随机分为缬沙坦组[予缬沙坦10mg/(kg.d)经口灌服]、红花黄色素粉针组(红花黄色素粉针62.5mg/(kg.d)腹腔注射)与模型组[予同等量0.9%氯化钠注射液腹腔注射],并设假手术组,每组10只大鼠。行HE、Masson染色观察实验大鼠梗阻侧肾脏组织形态学改变,并用免疫组化方法检测观察实验大鼠梗阻侧肾脏ColⅢ、TGFβ1在蛋白水平的表达。结果模型组及各治疗组与假手术组相比均呈现程度不同的病理损害,梗阻肾间质ColⅢ沉积明显增加,TGFβ1蛋白的表达也增加(P<0.05)。各治疗组与模型组比较,ColⅢ、TGFβ1表达明显减少(P<0.05)。2治疗组间比较差异无统计学意义(P>0.05)。结论输尿管梗阻后肾组织呈现程度不同的病理损害,肾间质ColⅢ沉积明显增加,这可能与TGFβ1表达增加有关。红花黄色素粉针及缬沙坦对其有一定的抑制作用。Objective To observe the effect of Safllor yellow powder injection on ColⅢ andTGFβ1 in Rats with tubulointerstitial fibrosis. Methods The model of renal obstruction was established by unilateral ureteral obstruction （UUO） in rats and then divided into four groups, including UUO group, UUOV group（treated with Valsartan 10 mg/（kg·d ））, UUOH group（treated with Safflor yellow powder injection 62.5 mg/（kg·d ））and shamoperation group. The morphological change of kidney tissue and protein expressions of Col Ⅲ and TGFβ1 in kidney tissue were observed. Results In UUO group and the other treatment groups there were different degrees of lesions of kidney compared with the shamoperation group. The expression of ColⅢ and TGFβ1 in kidney tissue were obviously increased （ P 〈 0.05）. Compared with UUO group, the pathological lesion and the expression of ColⅢ and TGFβ1 decreased significantly in the treatment groups（ P 〈 0.05）. The comparison between treatment groups showed that there was no significant difference （ P 〉 0.05）. Conclusion After unilateral ureteral obstruction the different degrees of pathological changes in kidney tissue were shown, which was related to the expression of TGFβ1. Safflor yellow powder injection and valsarten have an inhibitory effect.
