摘 要:目的:探讨不同氧张力对牙周膜细胞的增殖碱性磷酸酶活性(alkaline phosphatase activity,ALP)影响。方法:采用组织块法体外培养人牙周膜细胞,取第5代培养细胞分别在210mL/L(对照组)和10mL/L O2浓度(乏氧组)下培养1~3d,复氧组在相同的乏氧环境下培养1d和2d后分别移到常氧条件下继续培养1~2d后检测牙周膜细胞增殖能力与分泌碱性磷酸酶情况。结果:所有组别的细胞增殖率随培养时间呈依赖性的增高。乏氧组乏氧第2,3d与对照组有统计学差异。所有组别细胞的ALP活性随时间而降低,但乏氧第1,2,3天和复氧1d与对照组相比明显降低,差别具有统计学意义。结论:乏氧对牙周膜细胞的生物学有较大的影响,提示临床牙周炎及正畸所导致的缺氧环境对牙周膜细胞活性与功能有一定的影响。AIM: To investigate the effects of hypoxia and re -oxygenation on the proliferation and alkaline phosphatase expression of human periodontal ligament cells. METHODS: Human periodontal ligament cells (hPDLCs) were cultured under 21% and 1% O2 seperately for 1 - 3 days. In the re - oxygenation groups, cells were first incubated under the same conditions as the hypoxia groups for 24h and then returned to normal oxygenation conditions and cultured for 1 -2 additional days. RESULTS: Proliferation ratios is increased in all groups in a time- dependent manner. Proliferation ratios in the hypoxia group were significantly higher than the control group on the 2 and 3 days. Meanwhile, the ALP activity were decreased in all study samples with a statistical significant difference in both hypoxia and re - oxygenation groups. CONCLUSION : It is suggested that there is a significant effect of hypoxia on PDL cells on account of cell proliferation and ALP activity.
