超抗原SEA联合PML-RARα对外周血T细胞TCR Vβ亚家族基因表达的影响

摘　　要：目的探讨超抗原SEA联合PML-RARα多肽对外周血T细胞TCR Vβ亚家族基因表达的影响。方法分别将SEA、PML-RARα多肽以及SEA联合PML-RARα多肽与正常人外周血单个核细胞共同培养,20d后收集增殖细胞,利用RT-PCR及基因扫描技术分析诱导后增殖T细胞TCRVβ亚家族的利用和克隆性增殖的特点。结果单纯SEA诱导后,T细胞表达了11个TCR Vβ亚家族,且仍为多克隆增殖。单纯PML-RARα多肽诱导后,T细胞限制性表达8个Vβ亚家族,其中Vβ13、Vβ14表现出寡克隆或寡克隆趋势。PML-RARα多肽联合SEA共同诱导,其T细胞表达TCR Vβ亚家族依然呈明显的限制性,且Vβ13、Vβ14亚家族呈寡克隆、双克隆及寡克隆趋势。结论SEA能协同PML-RARα多肽诱导的T细胞克隆性活化与增殖。Objective To investigate the distribution and clonal expansion of TCR Vβ subfamily T cells induced by both PML-RARα peptide and Staphylococcal enterotoxin A （SEA） in vitro. Methods Peripheral blood mononuclear cells from healthy donors were cultured with PML-RARα peptide and SEA for 20 days. After induction, the distribution and clonal expansion of TCR Vβ subfamilies of T cells were examined by RT-PCR and GeneScan. Results Expression of 11 TCR Vβ subfamilies, which were polyclonal, were found after induction by SEA. Restricted expressions of TCR Vβ subfamilies were induced by PML-RARα peptide with or without SEA. Clonal expanded T cells could be identified in some Vβ subfamilies, which were preferentially used in Vβ13 and Vβ14 subfamilies. Conclusion The restricted expression and clonal expansions of the TCR Vβ subfamilies of T cells can be stimulated by PML-RARα peptide with superantigen SEA.
