摘 要:目的探讨不同神经毒对N27细胞Caspase-3的影响。方法使用不同的神经毒素1-甲基-4-苯基吡啶离子(1-methyl-4-phenylpyridinium,MPP+)和百草枯(Paraquat,PQ)处理N27细胞,四甲基偶氮唑盐[3-(4,5-dimethylthiazzol-2-y1)-2,5-diphenyl-tetrazdium Bromide,MTT]法检测细胞活性及代谢状态;RT-PCR检测凋亡相关基因Caspase-3的转录。结果经MPP+、百草枯作用后N27细胞活性下降,且其下降程度与MPP+、百草枯浓度的升高呈线性关系;MPP+、百草枯作用后N27细胞Caspase-3 mRNA表达水平升高。结论MPP+、百草枯引起N27细胞生长抑制,并可促进N27细胞中Caspase-3的表达和神经细胞凋亡的发生,神经细胞的凋亡可能是引发帕金森病的因素之一。Objective To investigate the influence of different neurotoxins on the expression of Caspase-3 in N27 cells, Methods N27 cells were incubated with different neurotoxins ( paraquat, MPP^+ ). The cell viability and metabolism state were measured by MTT assay. The Caspase-3 mRNA was quantified by reverse transcription polymerase chain reaction (RT-PCR). Result MPP^+ and paraquat induced the decreased cell viability and dopaminergic cell density depletion in the in N27 cell line by a dose-dependent manner. RT-PCR showed that Caspase-3 mRNA was increased in N27 cells after treated with paraquat or MPP^+. Conclusion The neurotoxins (paraquat and MPP^+ ) could increase the expression of Caspase-3 in N27 cells and contribute to the apoptosis of N27 cells. It indicated that the apoptosis of neuron cells might play an important role in the occurrence of Parkinson's disease.
