异博定注射液对慢性腹膜透析大鼠腹膜透析功能及腹膜形态的影响

摘　　要：目的探讨不同剂量的异搏定对高糖透析液作用下,腹膜透析大鼠模型腹膜间皮细胞的形态和功能的影响及它们之间的关系。方法48只SD大鼠随机分为对照组和异博定低、中、高剂量组4组,腹膜透析30 d后进行腹膜功能试验、CA125检测,同时对腹膜印片进行形态学分析。结果异博定中、高剂量组能明显升高透析大鼠透析液超滤量,与对照组相比较有统计学意义(P<0.01),且异博定各组对透析大鼠CA125、腹膜间皮细胞的表面积、细胞密度等均无明显影响,相互间比较无统计学意义(P>0.05)。结论透析液中加入中、高剂量的异搏定可增加腹膜透析鼠的超滤及溶质的清除,且长期使用不会导致对腹膜间皮细胞损伤,其作用机制可能与其减少腹膜间皮细胞肥大、保护腹膜间皮细胞、改善腹膜超滤功能有关。Objective To investigate the influence of different concentration of verapamil injection on the cell morphology and the peritoneal membrane function, and their relationship in peritoneal dialysis rat with infusion of high glucose dialysate. Methods Forty-eight SD rats were randomly divided into four groups： the simple dialysis group, the verapamil low dose group, the verapamil middle dose group and the verapamil high dose group. After treated with three different dose verapamil contained dialysate respectively for 30 days, the peritoneal membrane function was measured. The content of CA125 was tested and the rat peritoneal mesothelial cells（PMCs） morphology was analyzed from the cell imprint at the same time. Results The peritoneal membrane ultrafitration were significantly raised in the middle and high dose verapamil groups, com pared with the simple dialysis group（P〈0.01）. The content of CA125, the density of cell population and the surface area of PMCs were little changed in every dose verapamil groups （P〉 0.05）. Conclusion Not only the ultrafiltration volume and solute clearance could be enhanced significantly with middle and high dose verapamil injection added to high glucose dialysate in long-term use,but also mesothelial cell injury was prevented. It suggest thht verapamil injection could protect the mesothelial cell, reduce hypertrophy of the mesothelial cell and raise the membrane ultrafitration.
