消痰散结方干预胃癌组织增殖细胞核抗原(PCNA)表达影响的研究

摘　　要：目的:观察消痰散结方对胃癌组织增殖细胞核抗原(PCNA)表达的影响,探讨消痰散结方抑制胃癌细胞增殖的作用机制。方法:建立裸鼠人胃癌MKN-45原位移植模型,采用随机数字表法随机分成生理盐水组,消痰散结方低、中、高剂量组,5-氟尿嘧啶(5-Fu)组,每组8只。观察指标有:①观察各组荷瘤鼠肿瘤生长情况及抑瘤率;②观察消痰散结方干预胃癌组织PCNA蛋白表达的影响。结果:消痰散结方对裸鼠人胃癌原位移植瘤有较好的抑制肿瘤生长作用,消痰散结方低、中、高剂量组抑瘤率分别为36.62%、44.82%、58.11%,呈现出剂量依赖效应。结论:消痰散结方有较好的抑制肿瘤生长的作用,其抑瘤作用机制可能与下调肿瘤细胞PCNA蛋白的表达有关,进而抑制细胞增殖、分裂,控制胃癌的复发与转移。Objective:To discover the mechanism of Xiaotan Sanjie Decoction inhibiting gastric cancer cell proliferation by way of observing the expression of proliferating cell nuclear antigen (PCNA) on gastric cancer cell was detected after intervention of Xiaotan Sanjie Decoction. Method; The screened MKN-45 cancer cell was orthotopic transplanted in nude mice stomach to establish animal model, nude mice were divided into 5 groups(8 mice /every group):normal sodium group , low dose Xiaotan Sanjie group, mid dose Xiaotan Sanjie group, high dose Xiaotan Sanjie group and 5- Fu group at random. ①The growth of mice tumor , inhibition tumor ratio were observed in every group regularly. ②The expression of PCNA on gastric cancer cell was detected after inhibition of Xiaotan Sanjie Decoction. Result; xiaotanshanjie Decoction can better inhibit orthotopic transplantation tumor growth in nude mice stomach, the inhibition tumor ratio of low, mid、high dose Xiaotan Sanjie group was respectively 36. 62% ,44. 82% ,58. 11% , showing dose-dependent effect. Conclusion; Xiaotan Sanjie Decoction can better inhibit the tumor growth , the mechanism of Xiaotan Sanjie Decoction inhibiting the tumor may be related to reduction the expression of tumor cell PCNA protein, thereby inhibiting cell proliferation、separatism, controlling gastric cancer recurrence and metastasis.
