X线对脑胶质瘤细胞系增殖的影响

摘  要:为探讨不同X线处理对脑胶质瘤细胞增殖的影响,选用人脑星形细胞瘤SWO-38系,采用MTT法观察X线对胶质瘤细胞生长的影响,ABC法检测增殖细胞核抗原(PCNA)基因表达的变化。结果显示:①不同剂量(5,10,15,20,25Gy)X线对胶质瘤细胞生长具有明显的抑制作用,生长曲线低平,照射后48h的抑制率分别为33%、47%、51%、58%和66%;②不同剂量率(80,160,240,320,400cGy/min)X线照射后48h的抑制率分别为58%、54%、47%、25%和30%;③超分割照射后48h的抑制率为62%,而单次照射组为50%;④不同剂量(5,10,15,20,25Gy)X线照射后PCNA-标记率(LI)分别为91%,87%,81%,78%,69%,PCNA-LI与存活细胞OD值呈显著性正相关关系(P<0.05)。说明①SWO细胞生长曲线、抑制率与X线剂量、剂量率和照射方式之间密切相关;②PCNA-LI与存活细胞OD值呈正相关关系,PCNA是胶质瘤细胞增殖的标志之一。To investigate the effect of radiation by Xray on human glioma cells, modified MTT assay was adopted to observe the survival curves and inhibition rates of SWO cell lines after Xray radiation with the different modes. The results showed that: (1) There were significant inhibitive effects after different dose(5,10,15,25 Gy) Xray radiation on SWO cell lines. The inhibitive rates were 33%, 47%, 51%, 58% and 66% respectively after 48 hours of radiation. (2) The inhibitive rates were 58%, 54%, 47%, 25% and 30% respectively after 48 hours of the different doserate(80,160, 240, 320, 400cGy/min) radiation. (3) The inhibitive rates were 62% after hyperfractionational radiation, 50% for singlefractionation radiation. (4)PCNALI were respectively 91%, 87%, 78%, 69% after different dose(5,10, 15,20,25Gy)Xray radiation. PCNALI had significantly positive correlation with the OD value of survival cells (P<005). It was suggested that:(1)The survival curves and inhibition rates were closely related to the dose, doserate and radiation mode. (2)PCNALI was an important index of tumor cell susceptibility to radiation, and a symbol of S/G1 phrase in mitosis.
