摘 要:为探讨不同X线处理对脑胶质瘤细胞增殖的影响,选用人脑星形细胞瘤SWO-38系,采用MTT法观察X线对胶质瘤细胞生长的影响,ABC法检测增殖细胞核抗原(PCNA)基因表达的变化。结果显示:①不同剂量(5,10,15,20,25Gy)X线对胶质瘤细胞生长具有明显的抑制作用,生长曲线低平,照射后48h的抑制率分别为33%、47%、51%、58%和66%;②不同剂量率(80,160,240,320,400cGy/min)X线照射后48h的抑制率分别为58%、54%、47%、25%和30%;③超分割照射后48h的抑制率为62%,而单次照射组为50%;④不同剂量(5,10,15,20,25Gy)X线照射后PCNA-标记率(LI)分别为91%,87%,81%,78%,69%,PCNA-LI与存活细胞OD值呈显著性正相关关系(P<0.05)。说明①SWO细胞生长曲线、抑制率与X线剂量、剂量率和照射方式之间密切相关;②PCNA-LI与存活细胞OD值呈正相关关系,PCNA是胶质瘤细胞增殖的标志之一。To investigate the effect of radiation by Xray on human glioma cells, modified MTT assay was adopted to observe the survival curves and inhibition rates of SWO cell lines after Xray radiation with the different modes. The results showed that: (1) There were significant inhibitive effects after different dose(5,10,15,25 Gy) Xray radiation on SWO cell lines. The inhibitive rates were 33%, 47%, 51%, 58% and 66% respectively after 48 hours of radiation. (2) The inhibitive rates were 58%, 54%, 47%, 25% and 30% respectively after 48 hours of the different doserate(80,160, 240, 320, 400cGy/min) radiation. (3) The inhibitive rates were 62% after hyperfractionational radiation, 50% for singlefractionation radiation. (4)PCNALI were respectively 91%, 87%, 78%, 69% after different dose(5,10, 15,20,25Gy)Xray radiation. PCNALI had significantly positive correlation with the OD value of survival cells (P<005). It was suggested that:(1)The survival curves and inhibition rates were closely related to the dose, doserate and radiation mode. (2)PCNALI was an important index of tumor cell susceptibility to radiation, and a symbol of S/G1 phrase in mitosis.
