摘 要:研究了组合化学合成结构相似小分子多肽的毛细管区带电泳(CZE)和胶束电动色谱(MEKC)分析方法,均匀设计法选择出最佳缓冲溶液的组成,优化HPLC的流动相组成选择出最佳流动相。用最佳 MEKC、CZE和 HPLC法分别分析了10种多肽粗品并进行了比较,结果MEKC优于HPLC,HPLC优于CZE。To optimize the micellar electrokinetic chromatography (MEKC) method, the buffer composition, buffer pH, the content of organic additive and phosphate concentration are investigated and optimized by a statistic uniform design. The optimal MEKC buffer con- sists of 70 mmol/L of CTMAC, 20% acetonitrile and 70 mmol/L of phosphate at pH 2. 5. By the optimal MEKC method, ten crude peptides are analyzed and the results are compared with that obtained by the optimal CZE and HPLC. In view of the purity, migration time, column efficiency and the total number of peaks in each crude peptide, we might conclude that the MEKC method is superior to HPLC and HPLC superior to CZE.
