摘 要:目的:观察淀粉样蛋白(Aβ)1-42诱导的阿尔茨海默病(AD)大鼠模型经颅内注射Aβ1-42单克隆抗体(mAb)后其行为及脑组织病理的改变。方法:利用Aβ1-42蛋白片段诱导建立AD大鼠模型后,侧脑室内注入Aβ1-42 mAb,记录Morris水迷宫试验逃生时间作为行为学指标,同时取脑组织作切片染色,观察其病理改变。结果:AD大鼠模型颅内注射Aβ1-42 mAb后,水迷宫试验逃生时间明显缩短,表明其学习记忆能力明显改善;病理切片染色观察到脑组织内Aβ沉积斑块消失。结论:颅内注射Aβ1-42 mAb对AD动物模型有明显疗效。Objective: To observe the behavior and brain tissue variation after using amyloid peptide(Aβ-42 monoclonal antibody (mAb) cerebral injection in Alzheimer' s disease(AD) rat. Methods: AD rat can be created with the induction of Aβ1-42 peptide. After the injection of Aβ1-42 mAb in right side ventricle, escaping time of rats in Morris Labyrinth can be recorded as the action marker. Meanwhile, pathology assay should be done with its brain tissue. Results: After cerebral injection of Aβ1-42 mAb, the escaping time of AD rat in Morris Labyrinth decreased remarkably. That means its learning and memory ability obviously improved. The result of brain tissue pathology assay show that the Aβblot disappeared. Conclusion: Aβ1-42 mAb cerebral injection do have an obvious curative effect on AD rat.
