摘  要:目的通过体外实验研究槲寄生碱(mistletoe alkali,MA)对人结肠癌细胞(sw620 cells)增殖的抑制作用及其作用机制。方法采用cck-8试剂盒检测MA对sw620细胞增殖的作用。生长抑制率(%)=(A值对照孔-A值实验孔)/A值对照孔×100%。绘制其在不同浓度药物作用下的细胞生长曲线。采用PCNA免疫细胞化学染色方法观察MA作用后sw620细胞的表达情况。结果MA作用后,sw620细胞的增殖受到抑制,IC50为3.3μg/ml(P<0.001);加药组细胞生长明显缓慢;PCNA蛋白表达水平下降(P<0.01)。结论槲寄生碱(MA)对人结肠癌细胞(sw620 cells)增殖有显著的抑制作用,其机制可能是通过下调PCNA蛋白的表达实现的。Objective To study the inhibitory effects and mechanism of mistletoe alkali(MA) on the proliferation of sw620 cells (intestinal cancer cells ) through out-of-body experiments.Methods In virto,CCK-8 kit was used to measure the inhibitory effects of MA on proliferation of sw620 cells, inhibition ratio( % ) = ( Avalue of control groups - A value of experimental groups/A value of control groups ×100 %. The growth curve of sw620 cells with various concentration of MA was drawed. PCNA Immunocytochemicul stain was used to observe the expression of cells with MA. Results The proliferation of sw620 cells were inhibited by MA, IC50 value was 3.3 μg/ml ( P 〈 0.001) ;Growth curve showed the growth of sw620 cells of experimental groups were obviously slowly. PCNA Immunocytochemical stain showed that the expression of cells with MA was lower( P 〈0.01). Conclusion Mistletoe Alkali(MA) could obviously inhibit the proliferation of sw620 coils through down-regulating the expression of PCNA.
