摘  要:[目的]为进一步研究四氮杂大环配体生物学特性奠定基础,为其应用研究提供参考。[方法]以四膜虫为材料,应用流式细胞分析技术,探讨Zn(Ⅱ)-L、Co(Ⅱ)-L这2种配合物对四膜虫核DNA的损伤作用。[结果]经流式细胞仪检测,Zn(Ⅱ)-L、Co(Ⅱ)-L染毒2 h后,四膜虫DNA相对荧光强度出现明显差异。其中0.50 mg/ml Co(Ⅱ)-L引起的DNA的损伤最大,0.05 mg/ml Zn(Ⅱ)-L引起的损伤最小。随着DNA损伤的增大,相对荧光强度高的细胞数减少,相对荧光强度低的细胞数增多。流式细胞术能灵敏检测到DNA含量变化,可用于毒性检测的定性分析。[结论]Zn(Ⅱ)-L、Co(Ⅱ)-L均可引起四膜虫DNA的损伤而表现出毒性,Co(Ⅱ)-L的毒性大于Zn(Ⅱ)-L的毒性。[Objective] The aim was to lay the foundation for the further study on the biological characteristics of tetraazarmacrocycle ligands and provide the reference for the study of itsapplication. [Method]With the Tetrahymena thermophila as material, the damage effects of the 2 complexes of tetraazamacrocyele with Zn(Ⅱ) and Co(Ⅱ) on the nuclear DNA of T. thermophila were discussed by flow cytometry. [Result] The relative fluorescence intensity of T. thermophila DNA showed significant difference through detecting by flow cytormetry after Zn(Ⅱ)-L and Co(Ⅱ)-L were exposed to toxicant for 2 h. Among them, the DNA damage caused by 0.50 mg/ml Co(Ⅱ)-L was biggest and that caused by 0.05 mg/ml Zn(Ⅱ)-L was smallest, The number of cell with highly relative fluorescence intensity was decreased and that with lowly relative fluorescence intensity was increased with the increase of the DNA damage. The flow cytometry could detect the change of DNA content sensitively and could be used for the qualitative analysis of toxicity detection. [ Conclusion] Zn(Ⅱ)-L and Co(Ⅱ)-L all could cause the damage of T. thermophila DNA to exhibit the toxicity. The toxicity of Co(Ⅱ)-L was bigger than that of Zn(Ⅱ)-L.
