小鼠肠干细胞群对外源性草酸耐受性的研究

摘　　要：目的探索利用体外细胞培养技术构建的小鼠肠干细胞群对外源性草酸的耐受能力。方法利用细胞体外原代培养技术分离并培养小鼠肠干细胞群,将获得的细胞群种植于24孔板中;选取8孔生长状态较好的细胞,分别加入含终浓度为40、35、30、25、20、15、105、mmol/L草酸钠的细胞培养液,置入CO2恒温培养箱内培养72h后观察细胞状态。结果经反复实验可见草酸钠浓度为25 mmol/L的孔中,细胞生长速度减慢,出现少许空泡样改变;而浓度大于或等于30 mmol/L孔中,细胞出现大量空泡样改变,甚至崩解、死亡;草酸钠浓度小于或等于20 mmol/L的孔中细胞没有出现异常,生长状态较好。结论培养基中草酸浓度小于或等于20 mmol/L时,小鼠肠干细胞群能正常生长分化。Objective To study the tolerance of the intestinal stem cell population of the mouse to oxalate. Methods The intestinal stem cell population were isolated and cultured in a 24-pore plate. 8 pores of the cells whose status were well were selected, and the cell culture medium in which the concertration of the natrium oxalate was 40 mmol/L, 35 mmol/L, 30 mmol/L, 25 mmol/L, 20 mmol/L, 15 mmol/L, 10 mmol/L, and 5 mmol/L was used to culture the cells. The cells were put into constant temperature and cultured for 72 hours. Results The experiment had been conducted repeatedly. The speed of the cells growth decreased when the concentration of the natrium oxalate was 25 mmol/L; when the concentration was higher than 30 mmol/L, the vacuolus appeared and the cells died; but the cells cultured in culture medium in which the concertration of the natrium oxalate was or lower than 20 mmol/L had good status. COnClusioN When the concertration of the natrium oxalate in the culture medium is or lower than 20mmol/L, the intestinal stem cell population of the mouse can be well cultured.
