摘  要:目的探索建立兔自体血栓性脑梗死模型的方法,观察是否适合使用常规影像学设备研究。材料与方法家兔40只,对照组10只,实验组30只。颈部正中切口,穿刺颈外动脉,实验组向颈内动脉内注射自体静脉血栓,对照组仅注射生理盐水,3h后行CT灌注(CTP)成像、MR常规及扩散加权、扩散张量成像、数字减影血管造影(DSA)检查。观察手术侧大脑中动脉(MCA)供血区的血流灌注、MR信号及血管是否异常。结果实验组26只(86.67%)动物可见手术侧MCA区CT灌注异常、MRI异常信号及DSA上手术侧MCA闭塞,其中8只(30.77%,8/26)同时可见大脑前动脉供血区异常灌注及MR信号异常,病理学表现为Evans蓝(EB)异常染色,镜下可见急性脑梗死典型征象。对照组影像学及病理学均无异常。两组CTP灌注参数差异明显(P<0.01或0.05)。结论自体血栓性兔大脑中动脉梗死模型制作较简单,成功率较高,各种临床型影像学设备均能显示其异常。Objective To explore the method of rabbit model of embolic middle cerebral artery occlusion stroke with autologus clots and to investigate whether the model is suited to the study with conventional iamging equipment or not. Materials and Methods 40 rabbits were divided randomly into control group ( n = 10) and experimental group ( n = 30). Af- ter midline incision of the neck, external and interal carotid arteries ( ECA and ICA) were dissected. Autologus blood clots were injected into ICA through the incision of ECA for experimental rabbits, whereas only saline was infused for control group. CT perfnsion(CTP) ,conventional MR imaging, diffusion weighted imaging (DWI), diffusion tensor imaging( DTI), and digital subtraction angiography (DSA) were made 3 hours after the operation. We observed if there were abnormal blood perfusion,MR signal and angiographic signs to these rabbits. Results 26 experimental rabbits(26/30,86.67% ) were found abnormal perfusion and MR signal as well as the occlusion of middle cerbral artery,similiar abnomalities of pefusion and MR imaging were also detected in anterior cerebral artery perfusion area in 8 rabbits (30.77%, 8/26). Evans blue (BE) stairming and typical acute infarction signs were found on pathology for these 26 rabbits. No abnormal findings were found for control animals. There was obvious difference of CTP parameters between the two groups (P 〈 0.01 or P 〈 0.05 ). Conclusion The rabbit model of embolic middle cerebral artery occlusion stroke with autologus clots can be easily estab- lished, its abnormalities could be detected by conventional imaging equipments.
