摘  要:目的建立一种能很好模拟人体内生物学行为的胃癌裸小鼠原位移植和转移模型。方法采用人胃癌SGC-7901细胞株在裸鼠皮下经反复传5代形成实体瘤的完整组织块,建立人胃癌裸小鼠原位移植生长和转移模型16个。观察所建立的模型肿瘤生长状况、移植成功率和自发转移的发生率,常规H-E染色光镜下观察胃癌原位移植瘤、胃癌淋巴结转移和肝转移情况。电镜下观察肿瘤超微结构及免疫组化S-P法检测肿瘤诱导型环氧化酶-2(COX-2)。结果胃癌裸鼠原位移植块再次原位移植的原位成瘤率达100%(16/16),淋巴结广泛转移率为87.5%(14/16),肝转移发生率为75.0%(12/16),腹腔积液形成率为12.5%(2/16)。结论原位移植和转移模型均具有人胃癌自然生长过程的特点,具有人胃癌的生物学活性。Objective To develop a nude mouse model of human gastric cancer that can mimic its natural human biologic activities.Methods Human gastric cancer cell line SGC-7901 was cultured in nude mice repeatedly for 5 generations to get the subcutaneous solid tumor,which was then emulsified and orthotopicly inplanted into the anterior wall of the stomach in 16 nude mice.The tumor growth characters,tumor-take rate and metastatic rate were studied grossly,the transplanted tumor and its lymphatic and hepatic metastases,after routine H-E staining,the ultrastructures,by electron microscopy,and COX-,by immunohistochemical method.Results The tumor-take rate was 100%(16/16),the rates of metastasis to lymph nodes and liver were 87.5%(14/16) and 75.0%(12/16) respectively,the rate of ascites formation was 12.5%(2/16).Conclusion The model of human stomach cancer established is qualified,exhibiting natural growth characters and biological activities.
