摘　　要：目的探讨核转录子κB(NF-κB)decoy寡核苷酸(ODNs)对结肠癌SW480细胞Toll样受体4(TLR)4表达的影响。方法体外培养SW480细胞,LPS(10μg/L)刺激3 h后,用脂质体Lipofectin2000介导NF-κB decoyODNs转染6 h,RT-PCR法检测细胞的TLR4 mRNA。设对照组、LPS组、SODN组、Lipofectin2000组。结果转染NF-κB decoy ODN的SW480细胞的TLR4 mRNA表达明显高于对照组(P<0.01),但明显低于其他组(P均<0.01)。结论NF-κB decoy ODNs明显抑制SW480细胞TLR4的表达。Objective To study the effect of NF-κB decoy oligodeoxynucleotides （ODNs） on Toll-like receptor 4 （TLR4） expression in SW480 cells. Methods SW480 cells were cultured in vitro, were stimulated for 3 h with LPS（ 10 μg/L） , then added Lipofeetin2000 mediated NF-κB decoy ODNs for 6 h, TLR4 mRNA were examined by RT-PCR. The results were compared with control group, LPS group, scrambled ODNs group, Lipofectin2000 group. Results After SW480 cells were stimulated by LPS, TLR4 mRNA expression were significantly increased, the difference compared with control group was obvious（ P 〈 0.01 ）. After NF-κB decoy ODNs treating, TLR4 mRNA expression were significantly inhibited, but the Scrambled ODNs group and Lipofectin2000 group were no effect. Conclusion NF-κB decoy ODNs inhibite the expression of TLR4 in SW480 cells.
