摘  要:目的观察LIGHT、IFN-γ基因转染人肝癌细胞HepG2的凋亡及其Caspase-3和survivin的表达变化。方法用LIGHT、IFN-γ真核细胞表达质粒pcDNA4C-LIGHT-cDNA、pcDNA4C-IFN-γ-cDNA及其转化的E.coliJM-109感受态大肠杆菌,经质粒提取后,转染人肝癌细胞HepG2,同时设对照组(未转染)。分别于转染后12、24、48 h收集HepG2细胞。流式细胞仪检测细胞凋亡和Caspase-3、survivin。结果转染后细胞凋亡及Caspase-3表达量在对照组、LIGHT单转组、联合转染组呈逐渐递增趋势,survivin表达量呈递减趋势。结论LIGHT转染后能通过调节HepG2细胞Caspase-3及survivin的表达来发挥促细胞凋亡作用,INF-γ能增强LIGHT诱导的HepG2细胞凋亡,且显著上调其Caspase-3的表达。Objective To transfect the LIGHT and IFN-γ into HepG2 cells by eatiortie liposome mediated method and detect apoptosis and the expressions of survivin and Caspase-3 in transfered HepG2 cells. Methods The pcDNA4C-LIGHT-cDNA and pcDNA4C-IFN-γ-cDNA extracted from E. coil JM-109, were transfected into HepG2 cells mediated by cationic liposome. Non-transfection was as a control group. HepG2 cells were collected in 12, 24 and 48 h after transfection respectively. The apoptosis and the survivin and Caspase-3 were detected by FCM. Results The apoptosis and expression of Caspase-3 increased by degrees in all groups. The expression of survivin decreased by degrees in HGHT-mediated group and eotransfection group. Conclusions LIGHT has an effect on inducing the apoptosis of HepG2 cells by regulating the expression of Caspase-3 and survivin, IFN-γ can sensitize its effection signifieantlly.
