肺癌及癌旁组织中经典Wnt信号途径相关基因的甲基化状态观察

摘  要:目的观察人肺癌及癌旁组织中经典Wnt信号途径相关基因的甲基化状态,探讨其与肺癌发生的关系。方法应用巢式甲基化特异性聚合酶链反应(nMSP)检测25例新鲜肺癌组织及癌旁组织、11例存档肺癌组织、6例炎性假瘤的瘤旁正常肺组织以及肺腺癌A549细胞株的WIF-1、sFRP-1和DKK-3基因启动子区域的甲基化状态。结果6例正常肺组织未检测到甲基化基因。18例肺癌组织和10例癌旁组织中可检测到甲基化基因。在13例肺癌组织和2例癌旁组织可检测到2个以上甲基化基因。肺癌组织中WIF-1、sFRP-1和DKK-3三个基因的甲基化率分别为47.2%、36.1%、27.8%;肺腺癌A549细胞株中WIF-1和DKK-3基因呈甲基化状态,而sFRP-1基因呈未甲基化状态。结论肺癌组织中存在经典Wnt信号途径相关基因(WIF-1、sFRP-1、DKK-3)的甲基化。其导致的基因功能失活可能发生在肺癌形成的早期。Objective To detect the methylation of canonical Wnt signaling pathway-related genes in human lung carcinoma tissues and adjacent non-tumor area, and investigate the relationship between methylation in the promoter of canonical Wnt signaling pathway-related genes and carcinogenesis of lung carcinoma. Methods Tumor tissues and their adjacent non-tumor tissues of 25 patients with lung carcinoma, 11 paraffin imbedded tissues and normal lung tissues as well as lung adenecarcinoma cell line A549 were detected the CpG-island methylation in 3 canonical Wnt signaling pathway-related genes ( WIF-1, sFRP-1 and DKK-3) by nested methylation-specific polymerase chain reaction (nMSP). Results Methylation was not detected in any normal lung tissues, but was found in 18(72. 0% ) tumor tissues and 10(40. 0% ) adjacent non-tumor tissues from the patients with lung carcinoma. Methytation in 2 or more canonical Wnt signaling pathway genes was detected in 13 (52.0%) tumor samples and 2 (8.0%) adjacent tissues. Methylation rate of WIF-1, sFRP-1 and DKK- 3 detected were 47. 2%, 36. 1% and 27. 8%, respectively. WIF-1 and DKK-3 genes were methylated in lung adenocarcinoma cell line A549, but sFRP-1 was not. Conclusions The methylation of canonical Wnt signaling pathway-related genes is detected frequently in lung carcinoma tissues. A mechanism that leads to the dysregulation is likely to be involved in the early lung carcinogenesis process.
