卵巢上皮性肿瘤中SPARC的表达及意义

摘　　要：目的探讨富含半胱氨酸的分泌型酸性蛋白（SPARC）在卵巢上皮性肿瘤中的表达变化及其与卵巢上皮性癌生物学行为的关系。方法采用组织芯片及免疫组化技术检测20例良性卵巢上皮性肿瘤及63例原发性卵巢上皮性癌组织中的SPARC,同时对蛋白表达进行定位。结果原发性卵巢上皮性癌中SPARC表达水平为（4.19±2.28）分,明显低于良性卵巢上皮性肿瘤的（8.55±2.48）分,P〈0.05。卵巢上皮性癌中Ⅲ-Ⅳ期、低分化、有腹水及大网膜转移时SPARC较低,与对照者相比,P〈0.05。16例癌组织细胞核有表达,其余为细胞质表达。结论SPARC在卵巢上皮性癌中低表达;SPARC可能参与了卵巢上皮性癌的发生、发展、侵袭、转移过程。Objective To investigate the expression of secreted protein acidic and rich in cysteine （SPARC） in ovarian epithelial carcinoma and its relationship with pathologyical behavior of ovarian epithelial carcinoma. Methods The differential protein expression of SPARC in normal ovarian tissues and ovarian epithelial carcinoma was analyzed by tissue array technology and immunohistochemistry analysis and located using well characterized monoclonal antibodies. Results the expression of SPARC protein was significantly positive in ovarian epithelial carcinoma（98.41% ） compared to the normal ovarian tissues （P 〈 0. 05 ）. It was not associated with the tissue type,lymph node metastasis （P 〉 0. 05 ） ,but associated with the histological grade,aseites,eaul metastasis and the clinical stages of ovarian epithelial carcinoma （P 〈 0. 05）. SPARC located in cytoplasma except 16 cases. Conclusions SPARC is marked and may play an inhibited role in the development,growth and metastasis of ovarian epithelial carcinoma.
