摘 要:目的研究紫杉醇对人宫颈癌细胞株HCE1细胞凋亡率和细胞周期的影响。方法用紫杉醇处理人宫颈癌细胞株HCE1细胞,采用MTT比色法检测紫杉醇对宫颈癌细胞增殖的影响,确定紫杉醇抑制宫颈癌细胞的最佳浓度,通过流式细胞仪分析紫杉醇处理后宫颈癌细胞凋亡率和细胞周期的改变。结果紫杉醇对人宫颈癌细胞株HCE1细胞的增殖具有抑制作用,且随药物浓度的增加和作用时间延长,抑制作用越明显;低浓度紫杉醇只影响宫颈癌细胞的凋亡率不影响细胞周期,高浓度紫杉醇既影响宫颈癌细胞的凋亡率又影响细胞周期。结论紫杉醇能有效诱导人宫颈癌HCE1细胞的凋亡。[Objective]To explore apoptosis and cell cycle distribution of cervical carcinoma cell line HCE1 treated by Paelitaxel. [Methods] The effect of Pachtaxel on the proliferation of human cervical carcinoma cell line HCE1 was performed by MTT growth assay. The best concentration of Paclitaxel on cervical carcinoma ceils were de- termined .After the cells were treated with the concentration of Paclitaxel, the apoptosis assay and the cell cycle dis- tribution were analyzed by Flow cytometry. [Results] Paclitaxel could markly inhibit the proliferation of cervical carcinoma cell line HCE1. The inhibit effects were concentrated and time dependented. Low concentration Paclitaxel affected the apoptosis of cells and didn't affect the cell cycle distribution. High concentration Paclitaxel not only af- fected the apoptosis of ceils, but also affected the cell cycle distribution. [Conclusions] Paclitaxel can induce apop- tosis of cervical carcinoma cell line HCE1.
