摘  要:目的研究胚胎发育相关信号通路Sonichedgehog(Shh)对人胰腺癌细胞系PANC-1生物学行为的影响。方法试验组以cyclopamine阻断Shh信号通路,并设立对照,四唑蓝(MTT)比色试验检测Shh信号通路特异性抑制剂cyclopamine对胰腺癌细胞系PANC-1增殖的抑制作用;流式细胞术检测细胞增殖指数(PI)和凋亡指数(AI),Transwell侵袭小室检测两组PANC-1细胞侵袭能力的变化。结果cyclopamine对PANC-1细胞株增殖的抑制作用呈剂量和时间依赖性。cyclopamine作用后使胰腺癌细胞周期阻滞在G0/G1期,细胞凋亡增加;PANC-1的AI为(15.2±0.54),PI为(38.3±0.23)(P<0.05)。Transwell侵袭小室检测对照组穿透细胞数为(96±4)个,而实验组穿透数为(43±5)个。结论Shh信号分子对胰腺癌细胞的增殖起维持作用;通过特异性阻断该信号通路,可以抑制胰腺癌细胞增殖,促进细胞凋亡,并抑制癌细胞的侵袭能力。[Objective] To investigate the effect and mechanism of eyelopamine, the inhibitor of Sonic hedgehog signaling pathway, on the biological behaviour of pancreatic cancer cell line PANC-1. [Methods] The PANC-1 cells used in the experiment were euhured in vitro, and divided in experimental group and control group, and the MTI" assay was used to examine the anti-proliferative effect of cyclopamine. Flow eytometry (FCM) was used to ex- amine the effects of eyelopamine on the proliferation index (PI) and apoptosis index (AI) of PANC-1 cells. Transwell invasive cabin assay was used to test invasive ability of PANC-1. [Results] Cyelopamine produced time and con- centration dependent antiproliferative effects in PANC-1 cells. Cyclopamine could induce apoptosis and block cell cycle at GOG1 phase. The apoptosis index(A1) was (15.2±0.54), and proliferation index(PI) was (38.3±0.23) (P 〈0.05) Transwell invasive cabin assay showed that the number of penetration cells were (43±5) in experimental group, but were (96±4) in control group. [Conclusion] Shh signaling pathway is essential in the mataining of pancreatic cancer cell proliferation. Blocking shh pathway can inhibit the growth of pancreatic cancer cell and invasive ability, and in- duce apoptosis.
