摘  要:目的:探讨环氧化酶-2(cyclooxygenase-2,COX-2)基因及蛋白在乳腺癌中的表达及临床意义。方法:采用原位杂交和S-P免疫组织化学技术检测20例乳腺纤维腺瘤和52例乳腺癌中COX-2mRNA和蛋白的表达。结果:COX-2mRNA和蛋白在乳腺纤维腺瘤和乳腺癌组织中的阳性率分别为(20.0%、65.4%)、(15.0%、57.7%),差异有显著性(P<0.01);COX-2mRNA和蛋白表达之间差异无显著性(P>0.05)。COX-2mRNA表达与淋巴结转移和肿瘤大小有关(P<0.05),与年龄和组织学分级无关(P>0.05);COX-2蛋白表达与淋巴结转移有关(P<0.05),与年龄、肿瘤大小和组织学分级无关(P>0.05)。结论:(1)COX-2mRNA及蛋白在乳腺癌中表达上调,提示COX-2在乳腺癌的发生、发展中起重要作用。(2)COX-2与淋巴结转移正相关,提示COX-2可作为判断乳腺癌预后的标记物。Objective :To investigate the expressions of COX-2 gene and protein in breast carcinoma and analyze their relationship with clinical factors. Methods:The expression of COX-2 gene and protein were detected on breast fibroadenoma (20 cases) and breast carcinomas (52 cases) by in situ hybridization and immunoshistochemisty (SP method). Results :The positive rates of COX-2 mRNA and protein were (20.0%, 65.4%) and (15.0%, 57.7%)in breast carcinoma and breast fibroadenoma respectively.The expression of COX-2 in two groups had statistical difference(P〈0.01). The expressions of COX-2 mRNA and protein had no statistical difference(P〉0.05). COX- 2 mRNA expression was related to lymphatic metatasis and tunor size (P〈0.05),but not to age and histopathological grade (P〉0.05). COX-2 protein expression was related to lymphatic metatasis (P〈0.05), but not to age,tunor size and histopathological grade (P〉0.05). Conclusion: (1)Over-expression of COX-2 may play an important role in the pathogenesis and progression of breast carcinomas. (2) COX-2 expression is related to lymphatic metatasis ,suggesting COX-2 as a molecular marker of breast carcinoma.
