摘 要:目的研究喉鳞癌组织中CDK2表达在引起DNA异倍体发生过程中的作用。方法取手术中获得的50例喉鳞癌组织和30例声带息肉组织,用γ-微管蛋白抗体标记中心体,用免疫组织化学的方法检测CDK2激酶和γ-微管蛋白的表达;用流式细胞术检测喉鳞癌组织DNA倍体。结果在喉鳞癌组织中CDK2激酶和γ-微管蛋白阳性率表达[分别为68.0%(34/50)和78.0%(39/50)]都显著高于在声带息肉组织中(P<0.05)[分别为20.0%(6/30)和33.3%(10/30)],而且CDK2激酶的表达与γ-微管蛋白的表达具有相关性。21例CDK2表达阳性的喉鳞癌组织其DI为1.76±0.36;9例CDK2表达阴性的喉鳞癌组织其DI为1.05±0.07,CDK2阳性的喉鳞癌组织较阴性表达的组织DI增高(P<0.05)。结论喉鳞癌中CDK2过度表达导致肿瘤细胞DNA异倍体发生。在诊断和治疗喉鳞癌中,CDK2可能是一个有重要作用的指标。Objective To investigate the role of CDK2 in DNA ploidy in laryngeal squamous cell earcinoma(LSCC). Methods Fifty specimens of LSCC and 30 specimens of vocal cord polyp were obtained during operation. Centrosomes were detected by anti-γ tubulin antibodies. Immunohistochemical method was used to analyze the protein expression of CDK2. DNA ploidy was determined in using flow cytometry. Results The positive rates of CDK2 and γ-tubulin in LSCC were 68.0 % (34/50)and 78.0 % (39/ 50), respectively, significantly higher than those in the vocal cord polyp[ P 〈 0.05,20.0 % (6/30) and 33.3 % (10/30), respectively]. Expression of CDK2 in LSCC was positively related with expression of γ-tubulin. DNA index in LSCC of the positive expression of CDK2 ( 1.76 ± 0.36 ) was significantly higher than that in LSCC of the negative expression ( 1.05 ±0.07, P 〈 0.05). Conclusion The results indicate that overexpression of CDK2 may be strongly correlated with DNA heteroploid in LSCC;, and it may be an important marker in diagnosis and treatment of LSCC.
