摘  要:目的:研究电针对自由运行状态下小鼠视交叉上核(suprachiasmatic nucleus,SCN)内核心钟基因隐色素基因(cryptochromes gene,Cry)Cry1和Cry2表达的作用,为临床择时针灸效应规律的研究提供分子生物学依据。方法:选用昆明种小鼠,按体重随机分为空白组、捆绑组、电针组。电针组固定动物后,选"百会"和"长强"针刺,电压1-3V,疏密波,频率2/20Hz,时间15min。捆绑组只固定15min,不予其它特殊处理;空白组不予任何特殊处理。在小鼠进入自由运行(free running,FR)状态第3天于近似昼夜时间(circadian time,CT)6:00(即CT6)应用实时荧光定量RT-PCR方法观测电针对SCN内Cry1和Cry2mRNA表达情况的影响。结果:在CT6针刺,有降低Cry1基因表达的作用和下调Cry2基因表达的趋势。结论:针刺导引节律相位的作用可能与其下调SCN内Cry基因的表达、降低负反馈抑制作用有关。Objective: To research the effect of electro- acupuncture on gene Cry expression in suprachiasmatic nucleus ( SCN) of free - running mice, so as to provide evidences of molecular biology for chrono - acupuncture. Methods : Rats of Kunming species were randomly divided into blank group, binding group and electro - acupuncture group. Applying the method of quantitative RT- PCR, we observed the change of mRNA expression of Cry1 and Cry2 in SCN after acupuncturing "Baihui" (Du20) and "Changqiang" ( Du1 ) . Results: The mCry1 mRNA level was decreased and mCry2 mRNA level was tended tobe downregulated by electro- acupuncture in SCN at CT6. Conclusion: The effect of electro - acupuncture is possibly related to the down regulation of genetic expression of mRNA in Cry1 and Cry2 and the reduction of negative feedback inhibition.
