土贝母皂甙微囊栓塞肝癌后对VEGF、FGF2表达影响的实验研究

摘　　要：目的观察土贝母皂甙微囊栓塞剂对肝癌抑制和VEGF、FGF2表达的影响,评价土贝母微囊栓塞效果。方法将40只兔VX2肝癌模型随机分为3组,分别用生理盐水1ml肝动脉灌注、碘油1ml加1mg丝裂霉素混悬液肝动脉化疗栓塞以及土贝母微囊5.1mg/kg肝动脉栓塞。栓塞后1周制备光镜、电镜标本,免疫组织化学SP法检测肿瘤组织VEGF、FGF2表达情况。结果栓塞后土贝母微囊组、化疗栓塞组肿瘤体积有所缩小,对照组体积明显增大,3组间具有统计学差异(P<0.05)。对照组、化疗栓塞组、土贝母微囊组VEGF阳性率分别为83.3%、91.7%、91.7%,FGF2阳性率分别为41.7%、58.3%、50.0%,3组间无统计学差异(P>0.05)。结论兔VX2瘤行土贝母皂甙微囊栓塞后VEGF及FGF2表达未见明显增高,但肿瘤生长得到抑制,可以作为理想的末梢栓塞剂。Objective To study the effect on the expression of VEGF,FGF2 and the anti - tumor effect after embolization by tubeimoside mierocapsule in hepatic carcinoma. Methods 40 models of VX2 liver tumor rabbit were randomly divided into control group（ group A ） ,chemoembolization group（ group B） and tubeimoside microcapsule group（ group C ）. Three groups were treated respectively with 1 ml saline by infusion of hepatic arteria,lipiodol of 1 ml plus 1 mg Mitomycin（MMC） and tubeimoside microcapsules （5.1 mg/kg） by hepatic arterial embolism. The expression of VEGF and FGF2 was observed by the immunohistochemical method in routine paraffin - embeded section after embolized 1 week. Results The volumes of tumors were decreased in group B and C ,and increased in group A after treatment of tumors,there were statistic significances among them （ P 〈 0.05 ）. The possitive expression of VEGF and FGF2 was 83.3% ,91.7% ,91.7% and 41.7% , 58.3% ,50.0% in the three group, respectively, there were no statistic signifieances among them（ P 〉 0.05 ）. Conclusion The expression of VEGF and FGF2 is not significantly increased and the tumor＇ s volumes are decreased after embolization with tubeimoside microcapsules in VX2 tumors of rabbits. Tubeimoside microcapsule may be a potential peripheral embolization agent.
