喉鳞癌中eIF4E基因的检测及其临床意义

摘　　要：目的:探讨eIF4E在喉鳞癌(LSCC)中的表达及其与LSCC的关系。方法:采用免疫组织化学法检测52例蜡块LSCC组织、20例喉黏膜不典型增生组织、30例喉良性增生组织及10例正常喉黏膜组织中eIF4E基因蛋白的表达情况;用Western-blot法检测20例新鲜LSCC标本的癌中心区(TC)、过渡区(TR)及病理证实的无癌区(TF)中eIF4E的表达情况。结果:免疫组织化学法检测示:eIF4E在LSCC中表达的阳性率为100.0%,显著高于对照组。Western-blot法检测显示:eIF4E在TF、TR及TC的表达水平呈进行性增高。结论:过度表达的eIF4E是LSCC恶性改变及判断其恶性程度的一个分子标志,可为LSCC的基因治疗提供一定的理论基础。Objective：The expression of eIF4E were studied by detection their expression in laryngeal squamous cell carcinoma（LSCC）. Methods ：An immunohistochemical analysis with antibodies to eIF4E was performed on Fiftytwo specimens of the LSCC, Twenty specimens of dysplasia, Thirty specimens of hyperplasia and ten specimens of normal mueosa;Western-blot analysis was performed on tumor core,transition core,tumor-free core of twenty specimens of fresh LSCC using anti-eIF4E,antiserums. Results ：All fifty-two patients（100% ） overexpressed eIF4E in the LSCC,they were remarkably higher than that in the control groups. The expression of eIF4E protein level from tumor-free core ,transition core and tumor core of LSCC appeared progressive overexpression. Conclusion：Overexpression of eIF4E may be a molecular marker of malignant transformation and progression.
