摘  要:目的:采用辣椒辣素(capsaicin)刺激大鼠上矢状窦区(superior sagittal sinus,SSS)硬脑膜,观察中脑导水管周围灰质(PAG)c-fos蛋白的表达,以探讨化学刺激大鼠上矢状窦区硬脑膜的偏头痛模型的可行性。方法:以雄性SD大鼠为实验对象,在手术暴露其上矢状窦后,capsaicin刺激SSS区硬脑膜,应用免疫组织化学染色技术,观察中脑导水管周围灰质c-fos蛋白(Fos)表达的变化。结果:空白组、生理盐水对照组、capsaicin 1μM组,capsaicin 10μM组每张切片的Fos阳性神经元数分别为35.2±9.4、44.1±11、116.7±13.5和304.2±18.9。结论:辣椒辣素作为有效的化学伤害刺激激活了PAG的神经元,可以模拟偏头痛的发作过程。Objective: To investigate the feasibility of the migraine animal model induced by chemical stimulation on the superior sagittal sinus. Method: The rats were prepared by exposure to the superior sagittal sinus. The dura mater near the SSS was stimulated by capsaicin and c-fos (Fos) protein was detected in periagueductal grey matter (PAG) area in coronal brain sections by using standard avidinbiotin immunohistochemistry. Result: The numbers of Fos immunoreactive positive neurons in capsaicin 1 μM group and 10 μM group was 116.7 ± 13.5 and 304.2 ± 18.9 respectively. While in control group and vehicle group was 35.2 ± 9.4 and 44.1± 11 respectively. Conclusion : The PAG could be activated by the capsaicin chemical stimulation of the dura mater near the SSS. The migraine animal model by using chemical stimulation of capsaicin evoked the general process of migrain attacks.
