摘  要:目的研究α-突触核蛋白(α-SYN)在大鼠不同脑区线粒体的表达及对线粒体呼吸链复合体Ⅰ活性的影响。方法用免疫印迹方法检测不同脑区线粒体中α-SYN表达量的变化并测定α-SYN向分离线粒体的转运;通过观察340nm处NADH吸收值的变化测定线粒体呼吸链复合体Ⅰ的活性。结果线粒体α-SYN在不同脑区的表达量不同。人重组α-SYN与分离线粒体进行孵育后,以剂量依赖的方式转运到线粒体内。将不同浓度人重组α-SYN与线粒体孵育后,线粒体复合体Ⅰ活性下降,并且呈明显的量-效关系。结论线粒体α-SYN在不同脑区的表达量不同。线粒体的α-SYN有可能来源于胞质α-SYN向线粒体的转运,并调节复合体Ⅰ的活性。Objective To study the expression of mitochondrial ot-Synuclein (α-SYN) in different rat brain regions and its effect on complex I activity of mitochondrial cascade. Methods Immunoblot analysis was used to examine the expression level of mitochondrial α-SYN in different brain regions and the transportation of α-SYN into isolated mitochondria. The mitochondrial complex I activity was measured by checking the change of NADH maximal absorbance at 340 nm. Results α-SYN was found to localize in mitochondria throughout the rat brain, with the amount being varied in different brain regions. Incubation of recombinant α-SYN protein with the isolated mitochondria led to transportation of this protein into mitochondria. Incubation of α-SYN protein with isolated mitochon- dria decreased the complex I activity of in a dose-dependent manner. Conclusion The amount of mitochondrial α-SYN is varied in different brain regions. Mitochondrial α-SYN may come from the transportation of cytoplasmic α-SYN and may regulate the complex I activity of respiratory chain.
