摘 要:采用分步法SDS-PAGE和改良A-PAGE分析了49份小麦农家品种等资源的Glu-1,Glu-3,Gli-1位点基因组成.结果表明,Glu-1位点编码的HMW-GS组成共检测到14种图谱,其中占比例最大图谱(N,7+8,2+12)达44.9%,其次是(N,7+9,2+12)和(1,7+8,2+12)各占14.3%,(1,7+9,5+10)和(2*,7+9,2+12)各占4.1%,其余9种图谱各占2.0%;Glu-3位点编码的LMW-GS组成共检测到25种图谱,(a,j,c)占12.2%,(a,g,c)占10.2%,(new,g,c)占8.2%,(a,g,a),(a,j,a),(f,g,c)和(d,j,a)各占6.1%,(f,j,a),(d,g,c),(a,f,c)和(f,f,c)各占4.1%,其余14种图谱各占2.0%,Glu-A3位点新发现的基因图谱占14.3%;Gli-1位点编码的醇溶蛋白组成共检测到40种图谱,(a,f,l)和(f,l,i)各占6.1%,(f,l,a),(new,c,i),(b,g,l)和(l,l,a)各占4.1%,其它34种图谱各占2.0%,Gli-1位点新发现的基因图谱占16.3%;扬白麦、迟亚草、葫芦头麦、红秃头、红粒白芒、红火麦、矮孟牛IV中在Glu-3或Gli-1位点含有新发现的等位基因.The banding patterns of HMWgs, LMWgs and HMW gliadins of 49 wheat landracs were analyzed using two-step SDS-PAGE and a modified A-PAGE,respectively checked out according to the 20 standard cuhivars' patterns. 14 HWMgs band patterns were encoded by Glu-1 locus,the pattern with the highest frequency was ( N,7 + 8,2 + 12) (44.9%), next came ( N,7 + 9,2 + 12 ) and ( 1,7 + 8,2 + 12 ), taking up 14.3 %, respectively, ( 1, 7 + 9,5 + 1 0 ) and (2 ° , 7 + 9,2 + 12 ), taking up 4.1%, respectively and the rest 9 patterns took up 8.2%. A total of 25 LMWgs band patterns were encoded by Glu-3 locus and the pattern with the highest frequency was (a,j, c) taking up (12.2%) , next came (a,g,c) (taking up 10.2% ) ,(new,g,c) (taking up 8.2% ) ,(a,g,a) ,(a,j,a),(f,g,c) ,(d,j,a) , ( taking up 6.1% ) , ( f, j, a) , ( d, g, c ) , ( a, f, c ) , ( f, f, c ) ( taking up 4.1% ) , more than 5 alleles were at Glu-A locus and 16.3% of them were newly discovered, 6 alleles were at GIu-B 1 locus,3 alleles were at Glu-D 1 locus ; 40 HMW gliadin band patterns were encoded by Gli-1 locus,more than 7 alleles were at Gli-A 1 locus,and 12.2% of them were newly discovered, 10 alleles were at Gli-B 1 locus,7 alleles were at Gli-D 1 locus;There are 8 landraces such as Yangbaimai,Chiyacao,Huloutumai,Hongtutou, Honglibaimai, Xiaohongmang, Honghuomai, Aimengniu IV which has new alleles at Glu-3 or Gli-1 loci.
