摘  要:目的:研究他扎罗汀诱导体外培养的皮肤鳞状细胞癌细胞株Colo-16细胞的凋亡。方法:用不同浓度的他扎罗汀处理Colo-16细胞,流式细胞术检测Colo-16细胞早期凋亡率;Hoechst 33258荧光染色法观察凋亡细胞的形态学变化;印迹杂交法测定caspase 3蛋白表达水平;半定量逆转录聚合酶链反应法(RT-PCR)测定caspase 3 mRNA的表达。结果:他扎罗汀显著增加Colo-16细胞的早期凋亡,且Colo-16细胞早期凋亡率与他扎罗汀呈浓度依赖性(P<0.05);荧光染色可见细胞核染色质固缩、核破碎;活化的caspase 3表达显著增强,caspase 3 mRNA表达显著增强。结论:他扎罗汀在体外可诱导Colo-16细胞凋亡,caspase 3的活化参与此凋亡过程。Objective: To investigate the apoptosis induced by tazarotene on cultured cutaneous squamous cell carcinoma cell line Colo - 16. Methods: Colo - 16 cells were treated with different concentrations of tazarotene. Flow cytometry (FCM) was used to determine the cell apoptosis rate. The morphological change of apoptotic cells was observed by Hoechst 33258 fluorescent stmning. The expression of caspase 3 in Colo - 16 cells was detected by Western blotting and caspase 3 mRNA was detected by semi - quantity reverse transcription - polymerase chain reaction (RT- PCR). Results: Tazarotene significantly induced Colo - 16 cells apoptosis. The early apoptosis rate of the colo - 16 cells was in a drug concentration - dependent manner. Typical morphological changes of apoptosis such as condensed nuclei and nuclei fragments were discovered by fluorescent staining. The expressions of caspase 3 and caspase 3 mRNA were increased. Conclusion: Tazarotene can induce Colo - 16 cells apoptosis, caspase 3 activation may be involved in apoptosis of Colo - 16 cells.
