摘 要:目的观察同种异体骨髓基质细胞(BMSCs)经直接穿刺移植到急性肝损伤兔肝的病理改变。方法由雄性白兔的股骨和胫骨骨髓中提取BMSCs后分离、培养及传代3周,然后直接肝脏注射移植到D-氨基半乳糖(D-Gal)诱导的同种雄性兔急性肝损伤模型内。实验组(n=24)为直接肝脏内注射5mLBMSCs(约2×10^7个/mL);对照组(n=34)为直接肝脏内注射5mLD-hanks液。BMSCs移植后1、2、4周分别处死2组实验兔,观察肝脏组织病理学变化及肝脏坏死程度。参照评分标准,2位组织病理学专业人员采用双盲法对各实验动物的肝脏组织标本进行评分。结果与对照组比较,实验组的肝脏坏死程度减轻、肝细胞再生显著;2组组织学评分有统计学意义(P〈0.001)。结论BMSCs直接穿刺移植到D-Gal诱导的兔严重急性肝损伤的肝脏后可显著减轻肝脏坏死程度并刺激肝细胞再生,进而发挥强大的抗坏死作用。Objective To observe the histopathological changes in the liver after transplanting allogeneic bone marrow stromal cells (BMSCs) into liver of acute liver injury (ALI) via percutaneous needle injection. Methods BMSCs were derived from bone marrow obtained from femoral and tibial bones of male albino rabbits. BMSCs were separated, grown, and propagated in culture for 3 weeks. 58 syngenic male rabbits were reduced liver injury with D-galactosamine. 24 hours later, 5 mL (about 2 × 10^7 cells/mL) BMSCs or D-hanks solution were percutaneously injected into livers of BMSCs group (n=24) and control group (D-hanks solution group, n=34) respectively. The rabbits were killed at different time points (1 week, 2 week, 4 week). Histopathological changes and degrees of necrosis in the hepatic tissues were assessed by two histopathologists blinded to treated group and each other's results. Results The degree of liver necrosis was decreased and the proliferation of hepatocytes was significantly stimulated in the BMSCs group compared to the D-hanks group; the histopathological scores of the 2 groups were statistically significant (P〈0.001). Conclusion The transplantation of BMSCs have a significant anti-injurious ability.
