摘 要:目的:观察中药壳聚糖对肝癌和肺癌细胞株糖基转移酶表达的影响,探讨中药多糖在肿瘤预防和治疗的应用。方法:以浓度为50,100和200 mg/L的壳聚糖分别作用于肝癌细胞株SMMC-7721和肺癌细胞株A549,采用RT-PCR方法检测多肽:N-乙酰氨基半乳糖转移酶2(ppGalNAcT2)和β3GalT7的表达水平。结果:肝癌细胞株中pp-GalNAcT2和β3GalT7仅微弱表达,加药后作用不明显;肺癌细胞株中ppGalNAcT2和β3GalT7均有所表达,加入不同浓度的壳聚糖之后,ppGalNAcT2和β3GalT7的表达均受到抑制,且随着壳聚糖浓度的增加,抑制作用越明显。结论:肝癌细胞株SMMC-7721和肺癌细胞株A549 ppGalNAcT2和β3GalT7的表达不同,壳聚糖能够抑制肺癌细胞株A549中的糖基转移酶ppGalNAcT2和β3GalT7的表达,从而抑制了糖基化作用,对肺癌的预防和治疗具有一定的价值。Objective : To observe the effect of chitosan to the expression of different glycosyhransferases in liver cancer cell line and lung cancer cell line and to study the application of chitosan in preservation and treatment of tumor. Methods: Detect the expression of ppGalNAcT2 and β3GalT7 in SMMC-7721 cell line and A549 cell line by RT-PCR after chitosan added to the culture medium. The concentration of chitosan was 50,100,200 mg/L. Results: The mRNA expression of ppGalNAcT2 and β3GalT7 was in low lever in SMMC-7721 cell line and the action was not obvious. The mRNA expression of ppGalNAcT2 and β3GalT7 in A549 cell line was decreased after chitosan added to the culture medium. As the increased of concentration, the inhibition was stronger. Conclusion: This study showed that the expression of ppGalNAcT2 and β3GalT7 in SMMC-7721 cell line and A549 cell line was different. The glycosylation in A549 ceils was inhibited by the chitosan through inhibiting the expression of ppGalNAcT2 and β3GalT7. It was good for preservation and treatment of lung tumor.
