摘  要:目的探讨扇贝裙边糖胺聚糖(SS-GAG)对高糖所致ECV-304损伤的保护作用及其机理。方法体外培养生长良好的ECV-304,建立高糖诱导的细胞损伤模型,在高浓度葡萄糖(G lu55.5 mmol.L-1)状态下加入不同质量浓度SS-GAG,采用3H-TdR掺入法检测细胞增殖活性,特异性硝酸还原酶法检测细胞内NO、放射免疫法测定内皮素-1(ET-1)和血管紧张素Ⅱ(AⅡ)。结果高糖损伤组,细胞增殖活性受到明显抑制,细胞培养上清液ET-1、AⅡ的含量明显升高,与正常对照组比较差异有统计学意义(P<0.01,P<0.05)),细胞内NO分泌明显降低,与正常对照组比较差异有统计学意义(P<0.01)。SS-GAG明显减轻高糖对细胞损伤,细胞增殖活性、ET-1、AⅡ、NO均趋于正常,且随SS-GAG浓度增高保护效果越明显。结论高糖对ECV-304细胞具有明显的损伤作用,可导致ECV-304细胞形态和功能的紊乱,引起ET-1、AⅡ产生增多,NO分泌失调;SS-GAG通过调节ET、AⅡ和NO分泌,减轻高糖对ECV-304细胞的损伤,保护ECV-304。提示SS-GAG对防治心血管疾病和糖尿病(DM)血管病变及动脉粥样硬化(AS)的发生发展具有积极的意义。OBJECTIVE To explore the protective effect of scallop skirt-glycosaminoglycan(SS-GAG) on vascular endothelial cells(ECV-304) damaged by highly concentrated glucose and its mechanism.METHODS The ECV-304 was cultured,and the damage mode1 of endothelia1 cells was established by adding high doses of glucose(55.5 mmol·L-1) into the cultured cells.Under the condition of highly concentrated glucose,different doses of SS-GAG were added,then the cell proliferation activity was measured by the method of 3H-TdR,NO in cells was assayed by nitric acid reductase,and ET-1 and AⅡ were detected by radioimmunoassay.RESULTS Compared with the control group,cell proliferation was inhibited,the excretions of ET-1 and AⅡ in supernatant wereincreased obviously(P〈0.01),and the excretions of NO in cells were diminished obviously(P〈0.01).However,SS-GAG weakened the damaged effect of highly concentrated glucose on cell proliferation activity,ET-1 and AⅡ,NO activities were close to normal level.The protective effect was obviously under the higher concentration of SS-GAG.The obvious difference occurred between the group treated with SS-GAG and the groups treated with highly concentrate glucose(P〈0.01),but there was no difference between SS-GAG and normal control(P〉0.05).CONCLUSION Highly concentrate glucose had obvious damaged effect on ECV-304,which may induce cell morphology change,dysfunction,ET-1 and AⅡ and NO excretion unbalance.SS-GAG possessed obvious protective effect on damaged ECV-304 by modulating the secretion of ET-1 and AⅡ and NO.Our present study suggested that SS-GAG may have important effect on preventing and curing cardiovascular disease,diabetes and arthrosclerosis.
